健脾补肾方对小鼠Lewis肺癌抑制作用及对VEGF、CD44V6表达的影响

摘    要:目的观察健脾补肾方对小鼠Lewis肺癌抗肿瘤作用,并探索其抗肿瘤的相关机制。方法采用Lewis肺癌移植瘤C57BL/6小鼠模型观察健脾补肾方抗肿瘤作用,并采用免疫组化法检测肿瘤组织血管内皮细胞生长因子(VEGF)和细胞黏附因子第6变异株(CD44V6)蛋白的表达情况。结果①环磷酰胺组(CTX)、健脾补肾方低剂量组、健脾补肾方低剂量加环磷酰胺组、健脾补肾方高剂量加环磷酰胺组对移植性Lewis肺癌小鼠瘤体的抑制率为44%、17.7%、53.9%和72.4%,以健脾补肾方高剂量加环磷酰胺组抑制作用最佳;②与对照组相比,健脾补肾方低剂量组能增加荷瘤小鼠脾指数,有统计学意义(P<0.05)。低、高剂量健脾补肾方与CTX合用,能对CTX所致荷瘤小鼠脾脏萎缩有一定的拮抗作用;③健脾补肾方与环磷酰胺联合用药表现为两药作用相加;④健脾补肾方能下调Lewis肺癌小鼠肿瘤组织VEGF、CD44V6蛋白的表达水平。结论健脾补肾方对小鼠Lewis肺癌具有显著的抗肿瘤作用,以健脾补肾方高剂量加环磷酰胺组最佳,两药合用有相加作用,其作用机制可能与该药能降低肿瘤组织VEGF、CD44V6蛋白的表达水平有关。

Abstract:Objective To investigate the anti-tumor effect and mechanism of jian-pi-bu-shen-fang.Methods The mouse model of C57BL/6 bearing Lewis lung carcinoma was established to investigate anti-tumor effect of jian-pi-bu-shen-fang.The expressions of VEGF,CD44V6 protein were detected in tumor tissues by three-step immuno-histochemistry staining.Results ①Tumor growth in the mouse bearing lewis lung carcinoma was significantly inhibited in CTX group,low dose of jian-pi-bu-shen-fang group,low dose of jian-pi-bu-shen-fang + CTX group,high dose of jian-pi-bu-shen-fang +CTX group.The inhibitory rates were 44%,17.7%,53.9%和72.4%,respectively and the high dose of jian-pi-bu-shen-fang and CTX group displayed the most potent anti-tumor effect in the four groups.②The low dose of jian-pi-bu-shen-fang group significantly improved the spleen index of the mice bearing lewis lung carcinoma(P<0.05).③The combinative effect of jian-pi-bu-shen-fang+CTX showed an additive effect.④Jian-pi-bu-shen-fang down-regulated the expression of VEGF,CD44V6 protein in carcinoma tissues of the mice bearing lewis lung carcinoma.Conclusion Jian-pi-bu-shen-fang showed an effective anti-tumor effect and the effect of the high dose of jian-pi-bu-shen-fang + CTX group was optimum.Their combinative effect showed an additive effect.The molecular mechanism of anti-tumor activity could be concerned with down-regulation the VEGF,CD44V6 in tumor tissues.
