CYPIA1,GSTT1,GSTM1基因多态性与宁夏回族食管癌易感性的研究

摘    要:目的研究探讨CYP1A1,GSTT1,GSTM1基因多态性与宁夏回族食管癌易感性的关系。方法收集40例经术后病理确诊为食管癌的回族住院病例的石蜡组织切片和80例健康回族查体者外周血标本,树脂型DNA提取试剂盒提取DNA、PCR扩增特异性片段,酶切、电泳。结果食管癌组与对照组间年龄、性别、吸烟、饮酒差异无统计学意义(P>0.05);CYP1A1基因型的Msp1位点的3种基因型在食管癌组及对照组的分布差异无统计学意义(P>0.05);GSTT1(-)基因型(P<0.05)在食管癌组及对照组间差异有统计学意义。GSTM1(-)基因型(P>0.05)在食管癌组与对照组中的分布无统计学意义。T1(-)M1(+)、T1(+)M1(+)在食管癌组及对照组中的分布差异有统计学意义(P<0.05)。CYP1A1,GSTT1及GSTM1联合基因型在食管癌组及对照组中分布无统计学意义,但T1(-)M1(-)tc/cc联合基因在食管癌组呈现增高倾向(OR=2.212)。结论 CYP1A1的Msp1位点多态性不能单独导致回族食管癌的发生,GSTT1(-)基因型是宁夏回族食管癌发生的危险因素。GSTT1(+)GSTM1(+)在回族食管癌组及回族正常对照组中分布有明显差异,食管癌的发生可能与GSTT1及GSTM1基因型缺失有关。

Study of CTPIA1,GSTT1,GSTM1 polymorphisms and susceptibility on esophageal carcinoma in Ningxia Hui nationality

Abstract:Objective To investigate the relationship between CYP1A1,GSTT1,GSTM1 gene polymorphism and esophageal cancer susceptibility in Ningxia Hui nationality.Methods 40 samples form patients with esophageal cancer and 80 healthy control were collected.Both patients and healthy control are all local Hui people.The DNA samples were extracted form esophageal cancer paraffin-embedded tissue blocks by using resin-based DNA extraction kits.And genotypes of CYPIA1,GSTT1 and GSTM1 were analyzed.Results Both esophageal cancer group and control group were investigated by age,gender,smoking history as well as alcohol consumption,and results showed that no significant difference between two groups(P>0.05).MSP1 locus genotypes of the three genotypes in CYP1A1 in esophageal cancer group and control group were not significant distribution of difference(P>0.05);GSTT1(-) genotype in esophageal cancer group and control group were significantly different(P<0.05).GSTM1(-) genotype in the esophageal cancer group and control group were not significant differences in the distribution(P>0.05).T1(-) M1(+),T1(+) M1(+) in the esophageal cancer group and control group were different in the distribution(P<0.05).CYP1A1,GSTT1 and GSTM1 combined genotypes in esophageal cancer group and control group were not significant differences in the distribution.However,T1(-) M1(-) tc / cc group showed that combined genes in esophageal cancer might increase tendency(OR=2.212).Conclusion The Msp1 CYP1A1 polymorphism alone may not relate with the recurrence of esophageal cancer in Ningxia Hui nationality.However,GSTT1(-) genotype maybe one of the risk factors of esophageal cancer.The deletion of GSTT1 and GSTM1 genotypes maybe related with esophageal cancer in Ningxia Hui nationality.
