排铅食品调节铅中毒小鼠血、脑微量元素及抗氧化酶的剂量效应关系

摘    要:目的分析排铅食品影响铅中毒小鼠全血、脑组织中微量元素及抗氧化酶的剂量效应关系。方法选用75只18~22 g昆明种雄性小鼠,随机分为阴性对照组、醋酸铅模型组和排铅食品低、中、高剂量组。除阴性对照组外,其他组均自由饮用1.00 g.L-1醋酸铅水溶液染毒3,0 d后,排铅食品组按人体推荐摄入量0.60g.kg-1.d-1的51、0和20倍经口灌胃;阴性对照组和醋酸铅模型组灌胃去离子水。1个月后测全血、脑组织中微量元素(Ca、Pb、Fe、Cu、Zn)以及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)和丙二醛(MDA)的含量。结果铅中毒小鼠全血和脑组织中Pb、Ca、Cu以及MDA含量随着排铅食品剂量的增加而降低,Fe、Zn含量、SOD和GSH-PX活性随着排铅产品剂量的增加而增加。结论该排铅食品在排铅的同时,可有效调节全血和脑组织微量元素的平衡,增加抗氧化物酶活性,降低脂质过氧化物的产生。

Abstract:Objective To analyze the dose effect relationship between lead excreting food and trace elements,antioxidant enzymes in the whole blood and brain tissues in lead poisoning mice.Methods Seventy-five Kunming male mice with body weight of 18-22 g were randomly divided into five groups:negative control group,lead poisoning model group and low、medium and high dose lead excreting food treated groups.In addi-tion to the negative control group,other groups took water contained 1.00 g·L-1 of lead acetate freely for 30 days for lead exposure.The mice in treated groups were given the lead-excreting food by gavages,based on 5-,10-and 20-fold of the recommended dosage for human consumption(0.60 g/(kg BW·d)-1 while the mice in negative control group and model control group were given deionized water by gavages.The trace ele-ments and antioxidant enzymes content of whole blood and brain was determined a month later.Results Pb,Ca,Cu and MDA contents in whole blood and brain tissues of lead poisoning mice reduced with the lead excre-ting food dose increasing while Fe,Zn,SOD and GSH-PX content increased with the lead excreting food dose raise.Conclusion While the lead excreting food reduces lead content,it can efficiently adjust the balance of trace elements,increase the antioxidant enzyme activity and reduce lipid peroxide production in the blood and brain tissue.
