凋亡相关因子XIAP和Caspase-3在胃癌组织中的表达及其临床意义

目的探讨X连锁凋亡抑制蛋白(XIAP)和半胱天冬蛋白酶-3(Caspase-3)在胃癌组织中表达及其与临床病理特征的相关性。方法采用免疫组化PV-9000二步法检测70例胃癌组织、30例胃良性病变组织和30例癌旁正常组织中XIAP和Caspase-3蛋白的表达情况。结果胃癌组织中XIAP的阳性表达率明显高于胃良性病变组织和癌旁正常黏膜组织(P<0.05)。胃癌组织中Caspase-3的阳性率明显低于胃良性病变组织和癌旁正常黏膜组织(P<0.05),差异有统计学意义(P<0.05)。胃良性病变组织和癌旁正常黏膜组织中XIAP、Caspase-3的阳性率差异无统计学意义(P>0.05)。XIAP和Caspase-3的蛋白表达呈负相关(P<0.01),XIAP和Caspase-3的阳性表达率与胃癌患者的组织分化程度、浸润深度、淋巴结转移、临床分期相关(P<0.05),与年龄、性别、肿瘤大小无相关性(P>0.05)。结论随着胃癌恶性程度的增加,XIAP的表达增高,而Caspase-3的表达降低;XIAP和Caspase-3可能参与了胃癌的发生发展,是评估胃癌预后的潜在指标。

Expression and its clinical significance of XIAP and caspase-3 in gastric cancer tissues

Objective To study the expression of XIAP and caspase-3 in human gastric cancer tissues and its correlation with clinical and pathological characteristics.Methods Immunohistochemical staining(PV-9000 2-step plus)was applied to detect the expression of XIAP and Caspase-3 of gastric cancer tissues in 70 cases,30 cases of precancerous lesion tissues(including gastric polyp and gastriculcer)and 30 cases of adjacent normal gastric mucosa.Results XIAP protein positive expression rates in the gastric cancer tissues was higher than those in the precancerous lesion tissues and the adjacent normal gastric mucosa(P<0.001).Caspase-3 protein positive expression rates in the gastric cancer tissues was lower than those in the precancerous lesion tissues and the adjacent normal gastric mucosa(P<0.001).There was no significance related to XIAP and caspases-3 protein positive expression rates in precancerous lesion tissues and the adjacent normal gastric mucosa.The expression of XIAP was negatively with the expression of caspases-3(P<0.001).The expression of XIAP and Caspase-3 was closely correlated with tissue differentiation,tumor depth,lymph node metastasis and clinical stage of gastric cancer(P<0.05),but it was not correlated with sex,age and tumor size(P>0.05).Conclusion With the increase the malignant degree of gastric cancer,the XIAP expression increases,while expression of caspase-3 decreases,which may participate in the development and progression of gastric cancer.These proteins may serve as potential biomarkers for evaluating the prognosis of gastric cancer.
