摘    要:目的探索建立急性CO中毒迟发脑病(DEACMP)大鼠模型的有效方法,为其进一步研究提供基础。方法 72只SD大鼠分为空气组、CO I组、CO Ⅱ组,分别给予腹腔注射空气、纯品CO气体追加注射(CO I组首次注射99.99%CO纯品气体120mL.kg-1后,连续3次按首次剂量的1/2追加注射,间隔为4h;CO Ⅱ组追加注射剂量改为首次剂量的2/3,其余同CO I组)并动态监测动脉血HbCO,3周后用Morris水迷宫逃避潜伏期成绩筛选具有认知功能障碍的大鼠,即为DEACMP大鼠模型。抽取DEACMP组和空气组大鼠制备脑组织病理切片,观察脑组织结构及神经细胞变化。结果 CO Ⅱ组大鼠急性期动脉血HbCO维持在50.07%~60.77%;CO Ⅱ组逃避潜伏期高于CO I组及空气组,DEACMP发生率也高于其它两组;DEACMP组大鼠脑组织病理改变主要为神经元变性坏死,大脑白质广泛脱髓鞘。结论首次注射CO纯品气体(99.99%)120mL.kg-1,追加剂量上调为首剂2/3量的腹腔注射方法更适合制作DEACMP大鼠模型。

Abstract:Objective To explore a reliable method for establishing the rat model of delayed encephalopathy after acute carbon monoxide poisoning(DEACMP),and to offer a basis for futher research on DEACMP.Methods 72 SD rats were randomly divided into air group,CO I group and CO II group and continuous intraperitoneal injection were given with air and pure CO gas.Rats in CO I group were injected with 99.99% CO gas in 120mL·kg-1 for the first time and a half of primitive dose were injected after 4h,repeatly three times at a four-our interval.An additional injection at two-thirds of primitive dose were adopted in CO II group.HbCO in arterial blood were detected dynamically.DEACMP rats were selected through measuromg cognitive ability by the escape latency of searching the objective in the Morris watermaze after three weeks.The brain tissue of DEACMP and normal rats were taken for pathological analysis.Results HbCO in arterial blood of CO II group were maintained at 50.07%-60.77% during acute period.The escape latency of CO II group were significantly more than that in CO I group and air group.DEACMP incidence in CO II group was significantly higher than that in CO I group and air group.The pathological examination revealed apparent neuronal degeneration and necrosis,and white matter demyelination extensively in DEACMP rats.Conclusion The intraperitoneal CO injection method is a suitable method for establishing DEACMP model of rats.
