Graves′病患者经~(131)碘治疗后循环活性T细胞亚群的动态观察

摘    要:本文应用直接、间接双色免疫荧光染色,流体细胞测定仪技术,观察了经~(131)碘治疗的23例Graves′病患者的循环活性T细胞亚群的动态变化。~(131)碘治疗后的第一个月至第三个月,HLA-DR、Ta_1和UCHL_1活性T细胞亚群数目明显增加,以Vicia-villosa为标志的抗抑制细胞亚群较治疗前明显增加。实验结果提示:Graves′病患者经~(131)碘治疗后的抗甲状腺自身抗体浓度增加可能是由于T细胞的激活和抗抑制细胞亚群增加的共同结果。

Abstract:Radioiodine therapy for Graves'thyrotoxicosis produces a rise in thyroid autoantibodies in the first three months after treatment, but little is known of its effects on T cells. We have therefore followed the changes in T cells subsets in sequential samples from 23 patients with Graves' disease treated with radioiodine, using dual-colour flow cytometry.In the first month after treatment there was a significant rise in activated T cells, identified by the markers HLA-DR (Ia) and CDW26/Ta_1 (P<0.025 in both case). CD45RO-positive T cells, which are the prime population containig memory cells, also increased (P<0.025), but there was no change in CD45R-positiv, resting cells or in the CD4/CD8 (helper to cytotoxic/suppressor) ratio.Vicia villosabinding T cells, containing the contrasuppressor population, showed a more variable response,but the trend was to an overall increase from pre-treatment values (P<0.025). The change did not appear to be related to antithyroid drugs treatment,since they, were seen irrespective of whether patients continued such therapy. These results suggest that T cell activation and enhanced contrasuppressor activity may in part be responsible for the rise in autoantibodies after radioiodine therapy.
