高渗刺激对大鼠视上核神经元Fos蛋白和加压素表达的影响及年龄相关性研究

摘    要:为了解在正常状态和高渗刺激条件下,大鼠下丘脑视上核(SON)中神经元Fos蛋白和血管加压素(VP)表达水平的变化及年龄相关性,将青年和老年组大鼠分为高渗实验组和正常(对照)组,实验组取材前1.5h腹腔注射高渗盐水,应用免疫细胞化学方法,观察青年和老年组大鼠下丘脑视上核Fos蛋白和VP的表达,并用计算机图像分析仪对免疫反应结果进行处理。结果显示:实验组大鼠SON中Fos表达阳性细胞核的光密度较对照组明显增加;而老年大鼠SON中神经元VP的表达强度也明显高于青年且。推测老年组大鼠SON神经元Fos和VP表达的这种增龄性变化可能与老年时期血管调节的变化有关。

Abstract:To observe the age-related changes of Fos protein and vasopressin (VP) expression in the neurons of supraoptic nucleus (SON) after intraperitoneal saline, young and old age rats were divided into experimental and control groups. The experimental rats were intraperitonially injected with 1.5 mol/L saline (3.7 ml/250 g) 1.5 h before the rats were sacrificed. The expression of Fos protein and VP in supraoptic nucleus of young and old age rats were observed by using immunocytochemical method and the results were processed by computer image analysis system. Results indicated that the optical density of Fos positive nucleus in SON neurons of experimental rats were significantly increased in comparison with those of control groups, and the optical density of VP positive neurons in SON of old age rats were significantly increased in comparison with those of young age rats. It was concluded that the changes of Fos and VP in SON neurons of old age rats might be related with the changes of regulation of blood vessels in advancing age.
