环胞菌素A治疗再生障碍性贫血对骨髓细胞凋亡的影响

摘    要:目的探讨环胞菌素A(CsA)治疗对再生障碍性贫血(再障)患者骨髓单个核细胞Fas(CD95)抗原表达及调亡的影响作用.方法应用流式细胞仪测定24例再障患者CsA治疗前后骨髓单个核细胞(MNC)Fas表达率及凋亡率,其中急性再障7例,慢性再障17例;同时以20例正常人作对照.结果应用CsA治疗再障3个月时的总有效率58.3%.再障患者Fas表达率及凋亡率均高于正常对照(P<0.05);急性再障Fas表达率及凋亡率高于慢性再障,但差异无显著性(P<0.05).用CsA治疗后,再障患者骨髓单个核细胞凋亡率明显低于治疗前(P<0.05).结论CsA治疗可降低再障患者骨髓MNC的凋亡率,骨髓MNC的Fas表达率和凋亡率增高与再障发病有关.

Abstract:Objective: To evaluate the effects of cycloporin A (CsA) on the Fas antigen expression and apoptotic rates of bone marrow cells in aplastic anemia (AA) patients. Methods: The apoptotic rates and Fas antigen expression in the bone marrow mononuclear cells (MNC) in 24 AA patients including acute 7 and chronic 17, and 20 normal control were assayed by FACS, respectively. Results: The total effective rate was 58.3% in 3 month by using CsA. Fas antigen expression and apoptotic rates of AA patients were higher than that of controls (P<0.05). Fas antigen expression and apoptotic serious aplastic anemia (SAA) were higher than that of chronic aplastic anemia (CAA), but there was no significant difference (P<0.05). After the patients were treated with CsA, the percentage of apoptotic cells in AA patients was decreased (P<0.05). Conclusions: CsA can reduce the percentage of apoptosis of bone marrow cells in AA patients. Fas antigen expression and apoptotic increase in marrow MNCs of AA patients, which is related to pathological mechanism of AA.
