摘    要:目的探讨去铁胺(deferoxamine,DFX)对低氧处理的内皮细胞的保护作用。方法将ECV-304脐静脉内皮细胞复苏,分别加入不同浓度的DFX,在低氧环境下进行培养8h,计数各组的细胞生存率,测定各组上清液中一氧化氮(NO)、乳酸脱氢酶(LDH)和内皮素(ET)水平,同时进行对照组试验。结果实验组中不同浓度DFX的处理均使内皮细胞生存率高于对照组,差异有统计学意义(P〈0.05),LDH,ET的水平明显低于对照组(P〈O.05)。结论DFX提高了内皮细胞对长时间低氧的耐受,减少了内皮细胞在低氧后的坏死,对低氧的内皮细胞具有保护作用。

A Exploratory-study of the cytoprotection of deferoxamine on endotbelinm under bypoxia

Abstract:Objective This experiment aimed to study whether protective effect of DFX existed in endothelial cell. Methods ECV-304 human umbilical cell strain were cultured in vitro and divided into 6 groups according to the concentration of DFX in culture media. Endothelial survival rate were observed and NO,lactate de hydrogenase (LDH) and endothelin levels were measured to study the effect of DFX on endothelial cell. Results 24h pretreatment of DFX facilitated the synthesis of NO and LDH of endothelium( P〈0. 05 ). And the level of NO and LDH had positive correlation with the concentration of DFX( P〈0. 01 ). There were no obvious diferences existing in survival rate among the groups that accepted nomox/c treatment. The dfferences be- tween the level of NO,ET and LDH of pretreatment and normoxic-treatment were of no statistics significance. Cellular survival rate of each group after hypoxic treatment decreased obviously ( P 〈 O. 01 ) ; almost every DFX processed group but DFX 0. 025mg/ml group,had higher survival rate than the control (P〈0.'05);cellular survival rate had positive correlation with the concentraion of DFX. The levels of LDH and El" of each group increased(P〈0. 05) ; the levels of LDH and ET had negative corre- lation with cellular survival rate ( P〈0. 01 ). The level of NO of each group decreased (P〈0. 05 ) ;The level of NO had positive cor- relation with cell survival rate(P〈0. 01 ). Conclusion The predisposal of DFX had augmented endothelial hypoxic tolerance.
