骨唾液酸蛋白对乳腺癌细胞MCF-7侵袭转移能力的影响

摘    要:目的研究骨唾液酸蛋白(BSP)对乳腺癌细胞MCF-7侵袭转移能力的影响及机制。方法通过划痕试验、Transwell侵袭实验、细胞黏附实验和反转录-聚合酶链反应等观察BSP对乳腺癌细胞MCF-7侵袭转移能力的影响。结果BSP稳定高表达可使MCF-7细胞的侵袭能力明显增加。稳定高表达BSP的MCF-7-BSP细胞与胶原和纤维连接蛋白的黏附性增加,其基质金属蛋白酶(MMP)-2和MMP-9表达也增强。结论BSP可以促进MCF-7细胞侵袭转移能力,其可能的机制是通过增加细胞的黏附性、促进MMP-2和uPA表达等。

Abstract:Objective To investigate the influences of bone sialoprotein(BSP) on the invasive and metastatic abilities of MCF-7 cells and its mechanisms.Methods The influences of BSP on the invasive and metastatic abilities of MCF-7 cells and its mechanisms were investigated by scarification test,transwell invasion assay,cell adhesion assay,and RT-PCR.Results BSP overexpression could promote the invasive and metastatic abilities of MCF-7 cells.The MCF-7-BSP cells of BSP stable overexpression increased the adhesiveness with collagen and fibronectin,and the expression of MMP-2 and MMP-9 was also increased.Conclusion BSP can promote the invasive and metastatic abilities of MCF-7 cells.These biological function maybe correlate to its abilities of increasing the adhesiveness of the cells,and promoting the expression of MMP-2 and MMP-9.
