共刺激分子B7-CD28/CD152在过敏性哮喘中的作用

摘    要:目的 观察 5 0例过敏性哮喘患者不同时期共刺激分子CD2 8、CD15 2 (CTLA4)、CD80(B7 1)、CD86 (B7 2 )和外周血T淋巴细胞及其亚群CD3+ 、CD4+ 、CD8+ 、CD4+ /CD8+ 、CD2 1、CD2 5、HLA DR等变化 ,探讨这些分子在哮喘发病中的意义。方法 采用间、直接荧光标记流式细胞仪测定上述分子表达。结果 共刺激分子CD80、CD86在过敏性哮喘急性发作期明显上调 (P <0 0 5 ) ,CD2 8虽有升高趋势 ,但与健康对照组比较差异无显著性 (P >0 0 5 ) ;而CD15 2在哮喘急性发作期、缓解期均无变化。急性发作期、缓解期过敏性哮喘病人外周血CD3+ 、CD4+ 、CD8+ 、CD4+ /CD8+ 等T细胞亚群与对照组相比差异无显著性 ,但T细胞活化标记物HLA DR明显升高 (P <0 0 5 )。结论 共刺激分子B7 CD2 8/CD15 2可能参与了过敏性哮喘的发病过程 ;过敏性哮喘患者外周血T细胞及其亚群虽无比例失调但存在功能紊乱。

Abstract:Objective Costimulatory molecules(CD28,CD152,CD80,CD86),T cells and its subsets (CD3 +,CD4 +,CD8 +,CD21,CD25,HLA-DR,CD4 +/CD8 +)were investigated in 50 patients with allergic asthma.Methods Flow cytometry was used to evaluate the molecules expressed on the surface of peripheral blood mononuclear cells.Results The molecules of CD80,CD86 from patients with allergic asthma were significantly upregulated compared to those of normal controls(P<0.01).The T cells and its subsets had no changes compared to those of normal controls both in acute attack and stable asthmatics(P>0.05).HLA-DR,the activated marker of T cells,was increased in all stages of asthma.Conclusion The costimulatory molecules CD80,CD86 were upregulated and it is suggested that these molecules may involved in triggering acute asthma attack.The dysfunction of T cells obviously existed in asthma.
