沙苑子黄酮抗裸鼠人肝癌移植瘤的实验研究

摘    要:目的观察沙苑子黄酮(ACF)对人肝癌细胞SMMC7721裸鼠移植瘤生长的影响。方法30只裸小鼠建立人肝癌SMMC7721细胞皮下移植瘤模型后随机分为模型组、环磷酰胺(CTX,25mg·kg-1)组、ACF-1(400mg·kg-1)、ACF-2(200mg·kg-1)和ACF-3(100mg·kg-1)5组,给予相应药物,观察肿瘤生长曲线和移植瘤重量,计算肿瘤抑制率;免疫组化方法检测增殖细胞核抗原(proliferationcellnuclearantigen,PCNA)在肿瘤组织中的表达;光镜和电镜观察肿瘤组织病理变化。结果ACF-1组和ACF-2组裸鼠肿瘤体积和重量明显降低,ACF各给药组平均抑瘤率分别为68.20%、41.19%、26.79%;ACF-1组和ACF-2组肿瘤组织PCNA表达率明显减少(P<0.01,P<0.05);病理学观察结果,ACF可引起SMMC7721细胞坏死,诱导SMMC7721细胞凋亡。结论ACF可抑制SMMC7721裸鼠移植瘤的生长,其作用可能与下调肿瘤组织PCNA表达和诱导肿瘤细胞凋亡有关。

Abstract:Aim To observe the effect of flavonoids from Astragalus complanatus(ACF) on the growth of SMMC7721 human liver caner subcutaneous transplanting tumor in nude mice. Methods After the nude mice model of the subcutaneous transplanting tumor was established by human liver cancer SMMC7721, 30 cases were randomly divided into model group, CTX group,ACF-1 group,ACF-2 group and ACF-3 group(n=6,each),treated with saline, CTX and ACF respectively.The expression level of proliferation cell nuclear antigen (PCNA) in the tumor tissues was also investigated by immunohistochemical methods. The histopathological changes were observed by light microscopy and electromicroscopy. Results In comparison with that in the model group, the size and weight of tumors in ACF group were significantly decreased. The expression level of PCNA in tumors was decreased in both ACF-1 group and ACF-2 group, comparing with the control(P<0.01,P<0.05).The pathological alterations were observed. In ACF groups, degrees of necrosis in tumor tissues were markedly increased respectively compared with the model group and cell apoptosis can be observed. Conclusion ACF can inhibit apparently the growth of SMMC7721 human liver cancer subcutaneous transplanting tumor in nude mice. Its effect may be related to down-regulating the expression level of PCNA in tumors and inducing the apoptosis in SMMC7721 cells.
