醒脑静联合乌司他丁对重症急性胰腺炎大鼠脑损伤的保护作用*

摘    要:【摘要】目的 研究醒脑静联合乌司他丁对重症急性胰腺炎(SAP)大鼠脑损伤的保护作用。方法 120只雄性远交系大鼠随机分为12组:正常对照6 h组、24 h组,SAP 6 h组、24 h组,醒脑静处理6 h组、24 h组,乌司他丁处理6 h 组、24 h组,醒脑静联合乌司他丁处理6 h组、24 h组及阳性对照6 h组、24 h组;每组10只。分别在术后第6 h、24 h 观察神经功能评分。采集血液、胰腺组织、脑组织标本,观察血清淀粉酶水平、脑组织伊文蓝(EB)含量、胰腺组织及脑组织镜下病理学变化。应用放射免疫法检测血清肿瘤坏死因子(TNF)-α及白细胞介素(IL)-1β水平,应用逆转录-聚合酶链反应(RT-PCR)法检测脑组织TNF-α 信使核糖核酸(mRNA)及IL-1β mRNA的表达。结果 醒脑静联合乌司他丁处理24 h组血清淀粉酶水平、脑组织EB含量、胰腺组织及脑组织镜下病理学评分、血清TNF-α及IL-1β水平、脑组织TNF-α mRNA及IL-1β mRNA的表达水平均比SAP 24 h组显著降低,而神经功能评分则显著升高(P < 0.01)。醒脑静联合乌司他丁处理24 h组与阳性对照24 h组的上述各指标相比差异均无统计学意义(P﹥0.05)。结论 醒脑静联合乌司他丁对SAP大鼠脑损伤有较好的保护作用。

Protective Effects of Xingnaojing Combined with Ulinastatin on Brain Injury in Severe Acute Pancreatitis of Rats

Abstract:Abstract] Objective? To study the protective effects of xingnaojing combined with ulinastatin on brain injury in severe acute pancreatitis (SAP) of rats. Methods? A total of 120 male Sprague-Dawley rats were randomly divided into 12 groups including normal control 6-hour group,control 24-hour group, SAP 6-hour group, SAP 24-hour group, xingnaojing treated 6-hour group, xingnaojing treated 24-hour group,ulinastatin treated 6-hour group, ulinastatin treated 24-hour group, xingnaojing and combined ulinastatin treated 6-hour group, xingnaojing and combined ulinastatin treated 24-hour group,positive control 6-hour group and positive control 24-hour group. Each group included 10 rats. Neurobehavioral scores were observed, and a series of blood samples,pancreatic tissue and brain tissue samples were obtained at 6 and 24 hours after operation respectively. The serum amylase,brain tissue content of Evans blue (EB), the histopathological change of pancreas and brain were observed. The serum levels of tumor necrosis factor (TNF)-α and interleukin (IL)-1β were measured with radioimmunoassay (RIA). Levels of TNF-α mRNA and IL-1β mRNA in rat brain tissues were measured by RT-PCR. Results? Compared with SAP 24-hour group, the serum amylase,brain tissue content of EB, histopathological scores of rat pancreas and brain tissues,serum TNF-α and IL-1β, TNF-α mRNA and IL-1β mRNA in brain tissues were significantly decreased while neurobehavioral scores were significantly increased in xingnaojing combined ulinastatin treated 24-hour group (P < 0.01). However,there were no significant differences in all indexes between xingnaojing combined ulinastatin treated 24-hour group and positive control 24-hour group (P﹥0.05). Conclusion? Xingnaojing combined with ulinastatin plays a protective role in brain injury of SAP in rats.
