摘    要:目的]探讨气阴两虚型非小细胞肺癌肠道菌群、代谢组学和蛋白组学的基本特征。方法]选择气阴两虚型非小细胞肺癌患者13例作为患者组,选择健康志愿者15名作为健康对照组,收集两组粪便标本,用16S rDNA技术检测肠道微生态。采集两组血清标本,通过液相色谱-质谱联用技术比较两组代谢组学和蛋白质组学差异。最后通过WGCNA软件对多组学结果进行联合分析。结果]两组间存在6种显著差异菌,20种差异蛋白(上调5个,下调15个),11种差异代谢物(负离子模式下2个,正离子模式下9个),差异菌群、差异蛋白与差异代谢物间存在相关性,联合分析发现差异物质与癌症通路相关。结论]气阴两虚型非小细胞肺癌具有特异的肠道菌群、蛋白质和代谢物,与肿瘤的发生发展密切相关。

Multi-omics Analysis of Qi-Yin deficiency Syndrome of Non-small Cell Lung Cancer based on system Biology
Abstract:Objectives] To explore the basic characteristics of intestinal flora, metabonomics and proteomics of non-small cell lung cancer with deficiency of both qi and yin. Methods] 13 patients with non-small cell lung cancer with deficiency of both qi and yin were selected as patient group and 15 healthy volunteers were selected as healthy control group. Fecal samples of the two groups were collected and intestinal microecology was detected by 16s rDNA technique. The serum samples of the two groups were collected and the differences of metabonomics and proteomics between the two groups were compared by liquid chromatography-mass spectrometry (LC-MS). Finally, the joint analysis of multi-group learning results was carried out by WGCNA software. Results] There were 6 kinds of significantly different bacteria, 20 kinds of differential proteins (5 up-regulated and 15 down-regulated) and 11 kinds of differential metabolites (2 in negative ion mode and 9 in positive ion mode) between the two groups. there was a correlation between differential bacteria, differential proteins and differential metabolites. Joint analysis found that differential substances were related to cancer pathway. Conclusion] Non-small cell lung cancer with deficiency of both qi and yin has specific intestinal flora, proteins and metabolites, which are closely related to the occurrence and development of tumors.
