硒对大鼠血管平滑肌细胞表达细胞间黏附分子-1的影响

摘    要:目的:观察抗氧化剂亚硒酸钠(Se)对肿瘤坏死因子(TNF)-α诱导的大鼠血管平滑肌细胞(VSMC)细胞间黏附分子(ICAM)—1表达的影响,以探讨TNF—α介导单核细胞与VSMC黏附,致动脉粥样硬化及硒抗动脉粥样硬化的可能机制。方法:体外培养大鼠VSMC,分别为TNF—α(20ng/m1)及TNF-α(20ng/ml) Se(0.5-2.0μmol/L),孵有24h,采用细胞黏附实验、流式细胞术和RT—PCR测定ICAM—1蛋白及mRNA水平表达。结果:TNP—α可诱导VSMC ICAM—1表达增强;而硒可降低TNF—α诱导的ICAM—1mRNA水平及蛋白表达,且呈浓度依赖性。结论:抗氧化剂硒可抑制TNF—-α诱导的VSMC中ICAM—1的表达,减少单核细胞与VSMC黏附,这可能对延缓动脉粥样硬化的发生发展起一定作用。

Abstract:Objective:To observe the effect of selenium, an antioxidant, on TNF-a-induced expression of intercellular adhesion molecule-1( ICAM-1) in rat vascular smooth muscle cells (VSMC) and to investigate the mechanism of atherosclerosis related to adhesion between monocyte and VSMCs induced by TNF-a and of anti-atherosclerosis by selenium. Methods: Cell adhesion test was employed. Flow cytometric technique and RT-PCR were applied to study the protein and mRNA expression of ICAM-1. Results: Selenium inhibits the adhesion of monocytes to VSMSs and the expression of ICAM-1 in VSMCs induced by TNF-a in a dose-dependent manner. Conclusion: Selenium decreases monocytes adhesion to TNF-a-activated VSMCs by inhibiting the expression of ICAM-1 in VSMCs. This mechanism may contribute to the antiatherosclerotic role of selenium.
