次硫酸氢钠甲醛对小鼠海马神经胶质酸性蛋白表达的影响

摘    要:目的研究次硫酸氢钠甲醛对小鼠海马神经胶质酸性蛋白(GFAP)表达的影响。方法健康雄性昆明小鼠80只,随机分为对照组和次硫酸氢钠甲醛溶液0.4‰、0.8‰、1‰剂量实验组(n=20)。对照组小鼠喂食静置自来水;实验组小鼠分别代以0.4‰、0.8‰、1‰次硫酸氢钠甲醛溶液喂食,10 d和30 d时分别处死各组小鼠10只,取出大脑,Zambon i固定液固定,常规冰冻制片,免疫组织化学染色,光镜观察。结果10 d时实验组小鼠海马中GFAP阳性表达与对照组比较无显著差异(P>0.05),不同剂量组间也无显著差异(P>0.05);30 d时实验组小鼠海马中GFAP阳性表达明显增强(P<0.05),但不同剂量组间无显著差异差异(P>0.05)。结论次硫酸氢钠甲醛较长时间喂食对小鼠海马有损伤作用。

Abstract:Objective To study the influence of formaldehyde sulphoxylate on the expression of glial fibrillar acidic protein(GFAP) in mice hippocampus.Methods Eighty healthy male mice of Kunming genus divide into four groups at random:control group,0.4‰,0.8‰ and 1‰ test groups(n=20).Control group mice were fed with tap water.0.4‰,0.8‰ and 1‰ test groups were fed with 0.4‰,0.8‰,1‰ formaldehyde sulphoxylate solution.In the 10th and 30th day,the mice were killed,the cerebrum tissuce were took out,and fixed in the Zamboni fixation solution,conventionally produced frozen section and stained by immunohistochemistry.Results In the 10th day,there were no significant difference of the expression of GFAP among different dose group and control group.In the 30th day,the expression of(GFAP) in test group was increased experiment,but there was no difference among different dose groups.Conclusion It is harmful to mice Hippocampus that mice is chronically fed with formaldehyde sulphoxylate solution.
