金属蛋白酶-9在大鼠局灶脑缺血再灌注损伤中基因的表达变化

摘    要:目的探讨大鼠短暂大脑中动脉闭塞诱导的局灶脑缺血后金属蛋白酶-9(MMP-9)基因在不同时间的表达。方法制备大脑中动脉短暂缺血90min再灌注12h,24h,48h和7d的模型,用Western blotting和荧光定量PCR来检测MMP-9的蛋白和mRNA在不同时间的表达变化。结果 MMP-9蛋白和mRNA表达在缺血再灌注12h,24h,48h和7d均显著增加,其中48h为高峰期,7d开始下降。结论 MMP-9基因表达上调,提示MMP-9在局灶脑缺血再灌注损伤中起重要的作用。

Abstract:Objective To explore the expression of MMP-9 gene in different times after focal cerebral ischemia and reperfusion in the rat.Methods The middle cerebral artery occlusion 90 min and reperfusion 12 h、24 h、48h and 7d models were built in rat.The expressions of MMP-9 gene were observed by Western blotting and Real-time PCR.Result The expressions of MMP-9 protein and mRNA increased remarkably after ischemia and reperfusion 12 h、24 h、48h and 7d.It had a peak at 48h,and started to decrease at 7 d.Conclusion The expressions of MMP-9 gene increase in the focal cerebral ischemia and reperfusion in the rat,this indicates MMP-9 plays an important role in focal cerebral ischemia and reperfusion.
