川白芷香豆素有效部位的RP-HPLC指纹图谱研究

摘    要:目的:建立川白芷香豆素有效部位的高效液相色谱指纹图谱评价方法。方法:以川白芷香豆素有效部位为研究对象,应用RP-HPLC法,色谱柱为Aichrom Reliasfl C18(250mm×4.6mm,5μm),采用甲醇-水-四氢呋喃梯度洗脱模式,流速为1.0ml·min^-1,检测波长为300nm,建立了10批样品的RP-HPLC指纹图谱。结果:对10批样品的RP-HPLC指纹图谱进行分析,选定其中14个峰建立共有模式,并应用“中药色谱指纹图谱相似度评价系统”对样品进行了质量评价。结论:本文所建立的指纹图谱较全面地反映了川白芷香豆素类有效部位化学成分的特征,可用于川白芷香豆素有效部位的质量评价。

Abstract:Objective: To establish a way of RP-HPLC fingerprint analysis for the quality control of extract from Angelica dahurica (Fisch.ex Hoffm. ) Benth.et Hook.f. vat. formosana (Boiss. ) Shah et Yuan. Methods: PR-HPLC was adopted. Aichrom Reliasil C18 (250mm × 4.6mm, 5μm) was used, with mixture of methyl alcohol-water-tetrahydrofuran as mobile phase in a gradient mode. The flow rate was 1.0mL·min^-1. The wavelength was 300 nm. Fingerprints of ten batches of samples were determined. Results: A total of 14 common peaks in all the fingerprints of sample were confirmed to establish the mutual model.The sample quality was assessed by Similarity Evaluation System for Chromatographic Fingerprint. Conclusion: The method can be used to identify and evaluate conveniently the quality of extract from Angelica dahurica.
