溶血剂用量对新生儿血液分析中白细胞总数及分类的影响

目的探讨溶血剂用量在血液自动分析仪中对新生儿白细胞总数、分类计数、直方图的影响.方法对CELL DYN1600血细胞分析仪使用不同用量溶血剂,计数新生儿白细胞,观察其总数、分数计数及直方图的改变.结果随着溶血剂用量增加,白细胞总数及中性粒细胞绝对值降低,淋巴细胞绝对值和中间细胞绝对值增高,直方图恢复为正常曲线.结论新生儿处于生理缺氧状态,其红细胞数和血红蛋白都高(Hb＞200g/L).常规溶血剂用量不能将红细胞完全破坏,加大溶血剂用量,可以使白细胞正确分类,总数准确.

Effect of Lyse Dosage on the Total count and Differentiation of White Blood Cell of Newborns

Objective: Our purpose was to investigate the effect of lyse dosage on the total white cell count and differentiative count and histogram of newborns by automated hematology analyzer(AHA). Methods: Different dosage of lyse were used to count the white cells of newnborns. We observed the changes of total cell count and histogram of white cell by CELL DYN1600. Results: With the increase of the lyse dosage, the total white cell count and the absolute count of granulocyte decreased. The lymphocytes and mind range cells increased. The histogram of white cell recovered as normal. Conclusion: The quantity of red blood cell and haemoglobin is high(Hb>200g/L ) because the newborns are in physiological hypoxic state. Normal lyse dosage can not destroy the red blood cell completely .So white cell could be differentiated rightly by increasing lyse dosage,and obtain credible result in total count.
