塞来昔布对戊四氮点燃小鼠脑神经细胞Bcl-2和Bax蛋白表达的影响

摘    要:目的:观察塞来昔布对戊四氮点燃小鼠癫癎发作及脑神经细胞Bcl-2和Bax蛋白表达的影响。方法:C57BL/6小鼠30只随机分为对照组、模型组和给药组各10只,给药组予塞来昔布灌胃,对照组和模型组予生理盐水灌胃,模型组和给药组以戊四氮点燃法制备小鼠癫癎模型,通过行为学检测观察塞来昔布对小鼠癫癎发作的影响,免疫组织化学法测定塞来昔布对小鼠脑神经细胞Bcl-2和Bax蛋白表达的影响。结果:模型组小鼠脑组织Bcl-2和Bax蛋白阳性细胞数显著高于对照组(P0.01),Bcl-2/Bax比值显著低于对照组(P0.01);给药组Bax蛋白阳性细胞数显著低于模型组(P0.01),Bcl-2蛋白阳性细胞数及Bcl-2/Bax比值显著高于模型组(P0.01)。结论:塞来昔布能下调戊四氮点燃小鼠脑组织Bax蛋白表达水平,上调Bcl-2蛋白表达水平,提高Bcl-2/Bax比值。

Abstract:Objective:To explore the effects of celecoxib on the epileptic seizures and expression of Bcl-2 and Bax protein in neural cells of mouse brain following pentylenetetrazole-kindled epilepsy. Methods: Thirty C57BL/6 mice were randomly divided into control group (n=10),epilepsy group (n=10) and celecoxib treatment group (n=10). The effects of celecoxib on epileptic status in mice in different groups were investigated and the expression of Bcl-2 and Bax protein in brain cells of the mice were examined. Results: Relative to those in the control group,the Bcl-2 and Bax positive neural cells were significantly increased and the ratio of Bcl-2/Bax was significantly decreased in mice in the epilepsy group (P<0.01). The Bax positive neural cells was significantly decreased whereas the Bcl-2 positive neural cells and the ratio of Bcl-2/Bax were significantly increased in mice in the celecoxib treatment group when compared to those in the mice in the epilepsy group (P<0.01). Conclusion: Treating with celecoxib can down-regulate the expression of Bax protein,up-regulate the expression of Bcl-2 protein and increase the ratio of Bcl-2/Bax in brain of the mice following pentylenetetrazole-kindled epilepsy.
