Slug基因表达抑制对体外胰腺癌细胞生物行为的影响

摘    要:目的探讨siRNA干扰Slug基因表达对胰腺癌转移、血管生成和生长的影响。方法用脂质体转染法将表达siRNA-Slug的真核表达载体pGenesil-1-Slug-siRNA(pSlug-siRNA)和空载体pGenesil-1-Neg-siRNA(pNeg-siRNA)导入AS-PC-1细胞,G418筛选阳性细胞,获得稳定转染的阳性克隆。采用RT-PCR技术检测不同处理组细胞中Slug mRNA表达水平,Western-blot测定Slug蛋白表达水平。观察Slug沉默后及对体外胰腺癌细胞侵袭作用的影响;鸡胚绒毛尿囊膜(CAM)试验及CAM移植瘤实验检测Slug沉默对血管生成的影响,MTT比色法检测细胞存活率。结果pSlug-siRNA组细胞内Slug表达被有效沉默。重组细胞基底膜(Matrigel)侵袭实验显示,pSlug-siRNA组、对照组、pNeg-siRNA组穿透基底膜细胞数分别为(38±6.56)、(164±15.39)、(159.67±8.96)个/高倍镜(×200),差异分别有统计学意义(P0.05)。体外实验表明,AsPC-1细胞内Slug表达沉默后,血管密度明显降低,血管生成被抑制,细胞存活率明显降低。结论RNA干扰Slug基因抑制胰腺癌细胞生长、侵袭转移和血管生成,Slug可能成为胰腺癌基因治疗的新靶点。
