牛磺酸对四氯化碳染毒大鼠钙稳态失调的干预规律

摘    要:本研究中除设立正常对照组,四氯化碳(CCl4)对照组外,尚设立240mmol/L牛磺酸预投组(只在染毒前给予)和240mmol/L牛磺酸同投组(只在染毒同时给予)来对照考察80mmol/L和240mmol/L牛磺酸双投(染毒前及染毒过程中均给予)对500mg/KgCCl4经口染毒,2次/周,持续5周实验动物的干预作用。结果如下:1.240mmol/L牛磺酸双投对Ca2+-Mg2+-ATPase活性有部分保护作用,使其达到对照组的74.48%;对Cam活性亦有明显的保护作用,并使线粒体钙含量显著降低;80mmol/L双投,240mmol/L预投和240mmol/L同投均未有明确的干预作用。2.80mmol/L和240mmol/L牛磺酸双投作用存在明显差异。240mmol/L双投明显地防止了Ca2+-Mg2+-ATPase和Cam活性的抑制,对线粒体钙含量亦有显著的降低作用,而80mmol/L牛磺酸未见此效果。3.240mmol/L牛磺酸双投和保护作用明显,优于240mmol/L同投;240mmol/L牛磺酸预投的效果不明显。故研究提示应重视牛磺酸干预作用的时效性和量效性。

Abstract:Male SD rats were treated with 80,240mmol/L taurine in drinking water for 7 days prior to and simultaneously with subacute intoxicantion,CCl 4 dosing 500mg/kg was given two times/wk over 5 weeks.Groups were designed as normal control,CCl 4 treament,240mmol/L taurine pretreatment group(TPG,taurine treatment only prior to CCl 4 treatment) and 240mmol/L taurine simultaneous treatment group(TSG,taurine treatment simultanously with CCl 4 treatment) in contrast to 80,240mmol/L taurine double treatment groups(TDG).The result was shown as follows:(1) In 240mmol/L TDG there were partial protective effect on the activity of Ca 2 Mg 2 ATPase which was increased to 74 48% of that of control and complete protective effect on the activity of Cam,mitochondria calcium content were also decreased significantly; In 80mmol/L TDG,240mmol/L TPG and TSG,taurine all showed no antagonism against the disturbance of liver calcium homeostasis.(2) There were different significantly protective effects between 80 and 240mmol/L TDG.240mmol/L TDG could markedly prevent the activities of Ca 2 Mg 2 ATPase and Cam from inhibition and could increase of mitochomdria calcium content.However,80mmol/L TDG had no effect on calcium homeostasis.(3) The effects of 240mmol/L TDG was superior to that of 240mmol/L TSG and the effect of TPG werent't definite.Therefore,it suggest that it is very important to emphasis the dose effect and time effect relation of taurine.
