芪蛭益肺颗粒对慢性阻塞性肺疾病模型大鼠的治疗作用及机制研究

摘    要:目的:本研究观察了芪蛭益肺颗粒(QZYF)对慢性阻塞性肺疾病(COPD)大鼠的治疗作用,并探讨其作用机制。方法:将Wistar大鼠随机分为空白组、模型组、罗红霉素组、芪蛭益肺颗粒高剂量(2.912 g?kg-1)组、中剂量(1.456 g?kg-1)组、低剂量(0.728 g?kg-1)组,每组18只,雌雄各半。使用香烟烟雾暴露复合气道滴入脂多糖(LPS)的方法建立COPD大鼠模型。第61天测定各组大鼠肺功能指标。Elisa法检测大鼠血清及肺组织匀浆中白介素-8(IL-8)、白介素-13(IL-13)水平,生化法检测大鼠血清中羟脯氨酸(HYP)水平,并对肺组织进行HE及Masson染色。结果:肺功能检测结果显示,与模型组相比较,QZYF高、中、低剂量组FEV0.3、FEV0.3/FVC%、MVV均显著升高,RL显著降低(P<0.01或0.05)。QZYF高、中剂量组血清中IL-8含量显著降低(P<0.01,0.05),QZYF中、低剂量组血清中IL-13含量显著降低(P<0.01,0.05);QZYF中剂量组肺组织匀浆中IL-8、IL-13含量显著降低(P<0.05)。QZYF高、低剂量组血清中HYP含量显著降低(P<0.01,0.05)。HE染色结果显示,QZYF高、中、低剂量组肺组织病变均出现明显改善。Masson染色结果显示,QZYF高、中、低剂量组肺组织胶原纤维百分比均显著下降(P<0.05)。结论:芪蛭益肺颗粒可能是通过调控IL-8、IL-13等炎性因子的表达来减轻COPD模型大鼠肺组织炎性细胞浸润,降低肺组织胶原纤维沉积,缓解COPD模型大鼠肺组织气道重构,进而改善了COPD模型大鼠的肺功能。

Abstract:Aim: To investigate the treatment effects and mechanism of Qizhi yifei Granules on chronic obstructive pulmonary disease (COPD) rats. Methods: Wistar rats were randomly divided into 6 groups: normal control group, model group, roxithromycin group, and three dosage groups of Qizhi Yifei Granules with high (2.912 g?kg-1), medium (1.456 g?kg-1) and low (0.728 g?kg-1), 18 rats per group, half male and half female. A rat model of COPD was established by infusion of LPS with cigarette smoke exposure. At the end of the 61 days, all rats were sacrificed in order to determine 0.3% forced expiratory volume (FEV0.3), FEV0.3 / FVC%, maximum ventilation (MVV) and forced vital capacity (RL). The levels of IL-8 and IL-13 in rat serum and lung tissue homogenate and Serum hydroxyproline (HYP) levels in rat serum were measured. The lungs of rats in each group were subjected to HE and Masson staining. Results: FEV0.3, FEV0.3 / FVC% and MVV were significantly increased (P<0.01), and the RL was significantly decreased in the Qizhi Yifei Granules groups with high, middle and low dose (P<0.01, 0.05, 0.01). The results of Elisa assay showed the serum levels of IL-8 in high and middle dose of Qizhi Yifei Granules group were significantly decreased (P <0.01, 0.05), Serum levels of IL-13 in middle and low dose groups were significantly decreased (P <0.01, 0.05). The contents of IL-8 and IL-13 in the lung homogenate of the low dose group were significantly decreased (P <0.05). Biochemical tests showed that, the serum levels of HYP in the high and low dose groups were significantly decreased (P <0.01, 0.05).The results of HE staining showed that compared with the model group, the pathological changes of lung tissue in high, middle and low dose groups of Qizhi Yifei Granules were significantly improved. The results of Masson staining showed that Lung tissue collagen fiber area divided by the total area of lungs in high, medium and low doses of Qizhi Yifei Granules decreased significantly (P < 0.05). Conclusion: Qizhi Yifei Granules could reduce the infiltration of inflammatory cells in the lung tissue, improve the lung function of COPD model rats and reduce the deposition of collagen fibers in the lung tissue, ameliorate airway remodeling in lung tissue of COPD model rats of the model rats by regulating the expression of inflammatory factors such as IL-8 and IL-13, to protecting lung tissue of model rats finally.
