原代肠系膜动脉平滑肌细胞的分离培养

摘    要:取大鼠肠系膜动脉,用I型胶原酶消化,并进行体外培养,倒置相差显微镜观察ASMC 生长状态及特点;免疫组织化学进行细胞鉴定。大鼠肠系膜平滑肌细胞呈典型“峰-谷”样生长,该原代培养细胞对平滑肌特异性肌动蛋白表达阳性。传代细胞生长稳定,可为研究小血管平滑肌细胞特性提供一个较理想的模型。

Abstract:Mesenteric artery was obtained from rat and digested by type I collagenase, then cultured in vitro. The status and characteristics of mesenteric artery smooth muscle cells (MASMC) were observed ASMC with inverte phase contrast microscope and indentified by immunochemical. MASMC presented typical peak - valley -like growth and express positive for anti-alpha smooth muscle specific actin. SMC steady growth provides an available model for reseaching the specialities of small vessels.
