ER阳性乳腺癌中FOXM1和GRP78蛋白表达与临床病理因素及预后的相关性研究*

摘    要:目的:检测ER阳性乳腺癌组织中叉头框M1(forkhead boxM1,FOXM1)及葡萄糖调节蛋白78(glucose-regulated protein of 78kDa,GRP78)的表达水平,探讨其与临床各参数及预后的关系。方法:应用免疫组化技术检测FOXM1、GRP78在60例ER阳性乳腺癌中的表达,分析两者表达量的相关性,与临床病理参数的相关性及其对患者预后的影响。结果:FOXM1和GRP78在ER阳性乳腺癌癌组织中高表达,两者的表达呈正相关性;患者的无复发生存期(recurrence-free survival,RFS)与脉管侵犯、FOXM1、GRP78、Ki67表达、HER2表达有关,GRP78和FOXM1高表达均是ER阳性乳腺癌患者RFS的影响因素。结论:通过本研究发现FOXM1和GRP78在ER阳性乳腺癌癌组织中高表达,两者均影响ER阳性乳腺癌患者5年RFS时间。

Abstract:Objective:To explore the expressions of Glucose-regulated protein of 78kDa(GRP78) and Forkhead boxM1(FOXM1) protein in ER-positive breast cancer by immunohistochemistry technique, the relationship between their levels and clinical parameters as well as efficacy and prognosis was studied. Methods:The expressions of GRP78 and FOXM1 situation in the surgical specimens of ER-positive breast cancer of 60 cases were detected by IHC.The correlation of GRP78 and FOXM1 expressions and their effect on the prognosis of ER-positive breast cancer patient were analyzed. Results:The GRP78 and FOXM1 protein expression were over-expressed and positively correlated on the level of expression in ER-positive breast carcinoma tissue.Univariate Cox regression analysis showed that it was statistically significant between the patients of RFS in 5 years and vascular invasion, FOXM1, GRP78, Ki67 and HER-2 over-expressed. Kaplan-Meier survival curves and Log-rank test were used to analyze the 5-years of RFS time, showing that FOXM1 and GRP78 over-expressions were influential factors of efficacy respectivelyin ER-positive breast cancer tissue. Conclusions:It was found that FOXM1 and GRP78 were highly expressed in ER-positive breast cancer tissue, both of which affected the prognosis of ER-positive breast cancer patients.
