慢性乙型肝炎树突状细胞形态和吞噬功能的测定

目的 探讨慢性持续性HBV感染状态下树突状细胞（dendritic cells,DC）形态和功能的变化。方法 用密度梯度离心结合细胞贴壁和细胞因子共同培养的方法从外周血制备DC，观察其生长和形态；用HRP吞噬法观察DC成熟过程中吞噬能力的变化。结果 慢性乙型肝炎（CHB）患者DC数量较少，胞质突破起较少，形态不典型，在细胞发育分化成熟后仍保持较强的吞噬能力。结论 CHB患者的DC处于不成熟状态，可能与免疫耐受有关。

The Changes of Morphology and Phagocytosis of DCs from Chronic Hepatitis B Patients.

Aim To clarify the changes of dendritic cells(DC)in chronic hepatitis B infection.Method Peripheral mononuclear cells were isolated from peripheral blood with density gradient centrifugation,further cultured into dendritic cells with GM CSF and IL 4.The morphology of the cultures were microscopic photoed.The phagocytotic capacity of DC was studied by horseradish peroxidase phagocytosis.Results The morphology of DC from chronic hepatitis B(CHB) patients is atypical,with less cytoplasmal processes,not typical dendritic like.When the cells became mature,their phagocytotic capacity remained more potent than the normal DCs,Conclusion The DCs from CHB patients are immature,which is associated with the immunotolerance of chronic hepatitis B virus infection.
