沉香片剂小鼠口鼻吸入给药抗焦虑和催眠功效实验研究

目的:通过多种行为学检测,考察沉香片剂口鼻吸入给药对小鼠睡眠质量与焦虑状态的影响。方法:沉香片剂通过吸入给药模拟沉香传统给药方式,①利用焦虑三联小鼠行为模型,选择沉香片剂口鼻吸入给药评价其抗焦虑功效;②采用阈上和阈下剂量戊巴比妥钠协同催眠小鼠范式,选择沉香片剂口鼻吸入给药评价其催眠功效。结果:沉香片剂小剂量组能明显增加旷场实验中央区路程,沉香片剂小、中剂量组能明显延长旷场实验中央区停留时间;沉香片剂小、中、大剂量组能明显延长明暗箱明区停留时间;沉香片剂小、中、大剂量组能明显缩短阈上剂量戊巴比妥钠睡眠潜伏期、延长睡眠时间,沉香片剂中剂量组更能明显增多阈下剂量戊巴比妥钠动物睡眠只数。结论:沉香片剂口鼻吸入给药表现出明显抗焦虑活性,并能显著促进小鼠睡眠,对其自主活动无明显影响,同时证明了沉香口鼻吸入给药用于临床和亚临床抑郁症状治疗的合理性。

Experimental Study on Antianxiety and Hypnotic Effects of Aquilariae Lignum Resinatum Tablet on Mice by Oral and Nasal Inhalation

Objective:Through a variety of behavioral tests,to investigate the effect of Aquilariae Lignum Resinatum Tablet on sleep quality and anxiety state of mice by oral and nasal inhalation.Methods:The traditional administration method of Aquilariae Lignum Resinatum Tablet was simulated by inhalation of Aquilariae Lignum Resinatum,(1)Using the mice behavioral model open field test,elevated plus maze test and light-dark box test,the anti-anxiety effect of Aquilariae Lignum Resinatum Tablet was evaluated by oral and nasal inhalation;(2)The hypnotic effect of Aquilariae Lignum Resinatum Tablet was evaluated by using pentobarbital sodium hypnotic paradigm with suprathreshold and subthreshold dose. Results:The low dose group of Aquilariae Lignum Resinatum Tablet could significantly increase the central area distance in open-field test. The low and medium dose groups of Aquilariae Lignum Resinatum Tablet could significantly prolong the time in center area in open-field test. The low,medium and highdose groups of Aquilariae Lignum Resinatum Tablet could significantly prolong the time in the light box of light-dark box test. The low,medium and high dose groups of Aquilariae Lignum Resinatum Tablet could significantly shorten the sleep latency and prolong the sleep time in pentobarbital sodium hypnotic paradigm with suprathreshold dose. The mediumdose group could increase the number of sleeping animals in pentobarbital sodium hypnotic paradigm with subthreshold dose. Conclusions:The oral and nasal inhalation of Aquilariae Lignum Resinatum Tablet shows obvious anti-anxiety activity,and can significantly promote sleep quality in mice,and has no obvious effect on its autonomous activity,at the same time,it is proved that the oral and nasal administration of Aquilariae Lignum Resinatum is reasonable for the treatment of clinical and subclinical depressive symptoms.
