尼莫地平对人结肠癌细胞系增殖的影响

目的：探讨分析尼莫地平对人结肠癌细胞系（HCT）增殖的影响。方法采用不同剂量的尼莫地平作用于HCT细胞，通过细胞的生长动力学检测、细胞凋亡的流式细胞仪测定、瑞士-吉姆萨染色观察细胞形态。结果0．1～20．0μmol/L的尼莫地平加入HCT细胞培养基和对照组比较，12、24h后吸光度值均有明显差异，说明不同浓度尼莫地平对HCT细胞的增殖均有一定抑制作用，而在48h后，20．0μmol/L的尼莫地平作用于HCT细胞，抑制率可达35．67％。尼莫地平的浓度增高，G0～G1期的HCT细胞百分率逐渐升高，当尼莫地平浓度为10．0、20．0μmol/L，G0～G1期的百分率明显高于对照组的百分率，而S期的百分率则明显低于对照组的百分率，差异具有统计学意义（P＜0．05）。结论在一定剂量范围内，随着尼莫地平浓度的升高可以有效抑制HCT的增殖，为肿瘤的治疗提供新的方法。

Influence of nimodipine on the proliferation of human colon cancer cell line

Objective To investigate the influence of nimodipine on the proliferation of human colon cancer cell line（HCT cells） .Methods HCT cells were treated with different dosage of nimodipine .Then ,cell growth kinetics , apoptotic cells and cell morphology characteristics were analyzed .Results Compared with control group ,absorbance values of HCT cells ,treated by nimodipine with concentration of 0 .1-20 .0 μmol/L for 12 and 24 h ,were different . 48 h after treatment of nimodipine with concentration of 20 .0 μmol/L ,the inhibitory rate could reached 35 .67% . Proportion of cells at G0 -G1 stage increased with the elevation of the concentration of nimodipine .When the dosages of nimodipine were 10 .0 and 20 .0μmol/L ,the proportion of cells at G0 -G1 stage were significantly higher than con-trol group ,and that of cells at S stage was significantly lower （P〈0 .05） .Conclusion In a certain range of dosage ,el-evated concentration of nimodipine could effectively inhibit the proliferation of HCT cells ,which might be a new ap-proach for the treatment of cancer .
