苏黄止咳胶囊对大鼠慢阻肺模型NF-ΚB p65、TLR4及IAP1表达的影响

摘    要:目的:探讨苏黄止咳胶囊对大鼠慢阻肺模型的治疗作用及作用机制。方法:60只大鼠随机分为空白对照组、模型组、苏黄止咳胶囊低剂量组(0.36g/kg)、苏黄止咳胶囊中剂量组(0.72g/kg)、苏黄止咳胶囊高剂量组(1.44g/kg)、氨茶碱片组(0.01g/kg),除空白组外其余大鼠于实验第1天和第15天滴鼻注入0.2ml LPS(1 g/L)溶液。除第15天外,第2天至第28天每天烟熏30 min,造模后治疗4周,末次给药1h后使用动物呼吸功能测定仪测定大鼠最大呼气流量(PEP)、1s用力呼气容积(FEV1),采用HE染色和免疫组化染色方法观察各组大鼠肺部的病理改变。结果:苏黄止咳胶囊中、高剂量组可显著增加慢阻肺模型大鼠最大呼气流量(PEP)及1s用力呼气容积(FEV1);HE染色结果显示苏黄止咳胶囊中、高剂量组炎症分级显著降低,免疫组化结果显示苏黄止咳胶囊中、高剂量组大鼠肺脏细胞核因子/k基因结合核因子单克隆抗体(NF-KB p65)、Toll样受体4抗体(TLR4)表达显著降低;苏黄止咳胶囊各组大鼠肺脏凋亡抑制因子1抗体(IAP1)表达均显著降低。结论:苏黄止咳胶囊可能通过改善肺功能,降低慢阻肺模型大鼠肺脏NF-KB p65、IAP1和TLR4的表达,从而减轻慢阻肺模型大鼠肺组织损伤,起到对慢阻肺模型大鼠肺脏的保护作用。

Abstract:Objective:To study the therapeutic effect and mechanism of Suhuang Zhike Capsule on rat model of chronic obstructive pulmonary disease.Methods:Sixty rats were randomly divided into control group,model group,low dose group of Suhuang Zhike Capsule(0.36g/kg),middle dose group of Suhuang Zhike Capsule(0.72g/kg),high dose group of Suhuang Zhike Capsule(1.44 g/kg),aminophylline tablets group(0.01 g/kg).All rats except the blank group were intranasally injected with 0.2 ml of LPS(1 g/L)solution on the first and the 15th day of the experiment.After the model was treated for 4 weeks,the maximum expiratory flow(PEP)and the forced expiratory volume(FEV1)of the rats were measured using an animal respiratory function tester one hour after the last administration.The pathological changes of the lungs of each group were observed by HE staining and immunohistochemical staining.Results:The middle and high dose groups of Suhuang Zhike Capsule significantly increased the maximum expiratory flow(PEP)and forced expiratory volume(FEV1)in rats with chronic obstructive pulmonary disease.The results of HE staining showed that the inflammation grade of Suhuang Zhike capsule in middle and high dose group was significantly decreased,and the expression of NF KB p65(nuclear factor kappa-B p65)and TLR4(Toll like receptors 4)in lung of rats in middle and high dose group was significantly decreased;the expression of IAP1(inhibitor of apoptotic protein 1)in the each group of Suhuang Zhike Capsule was significantly decreased.Conclusion:Suhuang Zhike Capsule may reduce the expression of NF-KB p65,IAP1 and TLR4 in lungs of rats with chronic obstructive pulmonary disease by improving lung function,thereby alleviating lung injury in rats with chronic obstructive pulmonary disease.
