摘    要:目的 观察增龄对体外培养的新西兰兔角膜缘干细胞生物学特性的影响.方法 随机选取2月龄,1.5年龄,3年龄新西兰兔各5只,取其角膜缘组织,用酶消化法制成细胞悬液,以相同的密度种植,观察体外培养的角膜缘干细胞的生长情况,细胞免疫荧光鉴定,噻唑蓝(MTT)比色法检测其增殖活性,流式细胞术测细胞周期和细胞大小.结果 2月龄新西兰兔角膜缘干细胞较1.5年龄和3年龄增殖活性高,增殖期细胞和小细胞所占的比例大.结论 角膜缘干细胞的增殖能力随着年龄的增加而降低,其在上皮细胞中所占比例随年龄的增加而减小.

Abstract:Objective To investigate the effect of aging on the biological characteristics of limbal stem cells (LSCs)in vitro.Methods Selected 5 2-month-old, 1.5-year-old and 3-year-old New Zealand rabbits respectively,removed their epitheliums,dissociated into single cells by enzymatic digestion method,and seeded at the same density.The growth status was observed,identification was performed using immunofluorescence,proliferation was examined by MTT method and cell cycle,cell size were examined by flow cytometry. Results The proliferation, ratio of proliferating cells and small size cells of limbal stem cells in the 2-month-oldgroup were higher than those in the 1.5-year-old and 3-year-old groups.Conclusions The proliferation ability of limbal stem cells and ratio oflimbal stem cells in limbal epithelium decrease with aging.
