结直肠癌组织中DNA结合分化抑制蛋白1和MMP-9的表达及其与LMVD的相关性

摘    要:目的 探讨结直肠癌中DNA结合分化抑制蛋白1(Id-1)、基质金属蛋白酶9(MMP-9)和微淋巴管密度(LMVD)的表达、临床意义及相关性。方法 运用免疫组织化学法测定50例结直肠癌和50例癌旁结直肠组织中Id-1和MMP-9的表达,并以D-240为标志物,检测结直肠癌组织内LMVD值。结果 Id-1和MMP-9在结直肠癌组织中的阳性表达率分别为72.00%和78.00%,而在结直肠癌旁组织中的阳性率为24.00%和28.00%,差异均有统计学意义(均P<0.01)。结直肠癌中的LMVD值为(15.18±2.16),显著高于结直肠癌旁组织(5.24±1.09)(P<0.01);Id-1和MMP-9的表达及LMVD值与结直肠癌的浸润深度、TNM分期、肝转移、淋巴结转移、脉管浸润等均具有相关性(均P<0.01);癌组织中Id-1阳性表达组LMVD值明显高于阴性组(P<0.01);癌组织中MMP-9阳性组LMVD值明显高于阴性组(P<0.01);且结直肠癌组织中I d - 1 与MMP- 9 呈明显正相关性(P<0.01)。结论 Id-1和MMP-9的高表达与结直肠癌浸润和转移密切相关,两者表达上调可能是诱导结直肠癌微淋巴管生成及淋巴道转移的重要原因。

Abstract:Objective To explore the expressions of Id-1 and MMP-9 in colorectal carcinoma tissues and their correlation with lymphatic microvessel generation, and their clinical significance. Methods The expression of Id-1, MMP-9 and LMVD in colorectal adenocarcinoma tissues(50 cases) and colorectal pericancer tissues(50 cases) were examined by immunohislochemistry. Using D-240 mRNA as a marker, the LMVD value of colorectal adenocarcinoma tissues was measured. Results The positive expression of Id-1 and MMP-9 were 72.00% and 78% in colorectal adenocarcinoma tissues, which was significant higher than 24.00% and 28.00% in colorectal peri-cancer tissues(all P<0.01). The LMVD value was (15.18±2.16) in colorectal adenocarcinoma tissues markedly higher than (5.24±1.09) in colorectal peri-cancer tissues(P<0.01). Expression of Id-1, MMP-9 and LMVD value were correlated with the depth of tumor invasion, TNM stage, lymph node metastasis, vessel invasion, and liver metastasis(all P<0.01). The LMVD vlaue in Id-1 positive expression group was significantly higher than in Id-1 negative expression group (P<0.01). And the LMVD value in MMP-9 positive expression group was obviously higher than in MMP-9 negative expression group in colorectal adenocarcinoma tissues (P<0.01). The expression of Id-1 was obviously correlated with MMP-9 in colorectal adenocarcinoma tissues. Conclusion The high expression levels of Id-1 and MMP-9 have a close correlation with the invasion and metastasis of colorectal carcinoma. The up-regulation of them may play an important role in lymphatic microvessel generation and lymph node metastasis in colorectal carcinoma.
