外周血单核细胞自体吞噬相关基因表达与高血压颈动脉IMT的关系

摘    要:目的研究高血压患者外周血单核细胞的自体吞噬相关蛋白与颈动脉内膜中层厚度(IMT)的关系。方法 99例高血压患者分为对照组53例和IMT增厚组46例,用RT-PCR方法检测2组外周血单核细胞中自噬相关基因Beclin-1和LC3mRNA的表达,用Western blot技术检测相应蛋白的表达水平。结果 IMT增厚组自噬基因Beclin-1、LC3 ⅡmRNA(2.19±0.84)、(2.49±0.94)较对照组(1.00±0.37)、(1.02±0.44)表达升高(P0.01),相应蛋白IMT增厚组Beclin-1(1.80±0.67)、LC3-Ⅱ/LC3-Ⅰ(1.90±0.68)表达较对照组Beclin-1(1.07±0.32)、LC3-Ⅱ/LC3-Ⅰ(0.91±0.23)表达升高(P0.05)。结论高血压颈动脉IMT增厚患者外周血自噬相关蛋白Beclin-1和LC3的mRNA及相应蛋白表达增强,可能参与了颈动脉IMT增厚的进展。

Abstract:Objective To study the relationship between the autophagy related protein in peripheral blood mononuclear cells and intima-media thickness(IMT)of carotid artery in patients with hypertension.Methods Ninety-nine patients were selected as our subjects,who were divided into IMT increased group and control group.The expressions of Beclin-1 and LC3 mRNA of autophagy related genes in peripheral blood mononuclear cells were detected by RT-PCR and the expressions of relevant proteins were tested by Western blot.Results The expressions of gene Beclin-1 and LC3Ⅱ mRNA were(2.19±0.84)and(2.49±0.94)in IMT increased group,significantly higher than(1.00±0.37)and(1.02±0.44)in control group(P<0.01).Accordingly,the expressions of relevant proteins were(1.80±0.67)and(1.90±0.68)in IMT increased group,obviously higher than(1.07±0.32)and(0.91±0.23)in control group(P<0.05).Conclusion The expressions of autophagy related genes and corresponding proteins in peripheral blood mononuclear cells are tremendously increased in hypertension patients with thickening IMT,and may contribute to the development of thickening IMT.
