摘    要:目的:研究红芪多糖(HPS)的脱蛋白、脱色工艺。方法:采用L9(34)正交试验和单因素试验分别优选Sevage法脱蛋白工艺与过氧化氢脱色素工艺。结果:Sevage法脱蛋白工艺为:样液与试剂的体积比为1∶1,氯仿与正丁醇的体积比为4∶1,振摇30min,静置20min。过氧化氢脱色素条件是脱色时间为2~4h,用量为5~20ml过氧化氢每50ml样液。结论:优化后的条件适合于红芪多糖的脱蛋白、脱色素。

Abstract:Objective: To study the process of removing proteins and depigmentation of polysaccharide from Radix Hedysari. Methods: Used L9(34) orthogonal test and single factor experiment optimize the technological of removing proteins by Sevag and depigmentation by H2O2. Results: The optimal technical conditions of deproteinization of Sevag were: the volume ratio of polysaccharides solution-Sevag reagent was 1:1,the volume ratio of chloroform-n-butanol was 4:1,shaked for 30 minutes,and put for 20 minutes. The process of depigmentation of H2O2 was: the time of depigmentation was 2 to 4 h, the adding amount of H2O2 was 5 to 20 ml per 50 ml sample solution. Conclusion: The optimal process of removing proteins and depigmentation is fit for polysaccharide from Radix Hedysari.
