| 摘 要： | 目的 介绍一种快速原代培养、分离、纯化视神经星形胶质细胞的方法。方法 对P2期小鼠的视神经进行组织块原代培养，振荡法分离纯化，形态学观察，免疫组织化学染色。结果 培养的星形细胞纯度高，GFAP染色显示98%阳性。结论 组织块原代培养，振荡法分离纯化视神经的星形胶质细胞，对细胞的损伤小、纯度高，是快速得到该细胞的理想途径。

| Abstract: | Objective To introduce a method for condition primary culture of astrocyte of optic nerve.Methods Conditioned medium was made of GGF-BF ,and astrocyte of optic nerve was cultured and purified by shaking.Identification was made by means of microscope observation and immunohistochemistry.Results Astrocyte of rat optic nerve was cultured and purified,which showed positive reaction to glial fibrillary acidic protein(GFAP). Conclusion Conditioned primary culture of rat optic nerve, purified through shaking,is a reliable and quick method. |
