人骨髓间充质干细胞Toll样受体3和4活化对IL-10、IL-12和IL-6分泌的影响

目的：研究人骨髓间充质干细胞（BM—MSCs）中Toll样受体（TLR）3和4的活化对IL-10、IL-12和IL-6分泌的影响。方法：用RT—PCR法检测体外培养的BM—MSCs中是否存在TLR3和TLR4的表达；用不同剂量的聚肌胞和脂多糖分别进行诱导后，用ELISA法检测培养上清中IL-10、IL-12和IL-6的变化。结果：BM—MSCs中有较高水平的TLR3和TLR4mRNA表达。聚肌胞可明显促进IL-6分泌（P〈0．05），且0．025g／L组明显强于0．0025g／L组（P〈0．01）；明显抑制IL-10分泌（P〈0．01），且0．25g／L组明显强于0．025g／L组（P〈0．05）；对IL-12的分泌无明显影响。脂多糖可明显促进IL-6的分泌（P〈0．01），而对IL-12和IL—10的分泌无明显影响。结论：BM—MSCs中有较高水平的TLR3和TLR4表达。TLR3活化影响BM—MSCs对IL-6和IL-10的分泌，TLR4活化影响IL-6的分泌，TLR3或TLR4活化均不影响IL-12的分泌。

Effect of activation of toll - like receptor 3 and 4 in human bone marrow - derived mesenchymal stem cells on secretion of IL - 10, IL - 12 and IL - 6

Objective： To study the effect of activation of toll - like receptor 3 and 4 （ TLR 3 and TLR4） in hu- man bone marrow - derived mesenchymal stem cells （ BM - MSCs） on secretion of IL - 10, IL - 12 and IL - 6. Methods： The RT - PCR method was used to detect whether TLR 3 and 4 were expressed in BM - MSCs cultivated in vitro. After the BM - MSCs were incubated with different doses of POLY or lipopolysaccharide （LPS） , the super- natants were collected and the contents of IL- 10, IL- 12 and IL- 6 were detected by ELISA. Results：Over- ex- pressions of TLR3 and TLR4 mRNA were found in BM - MSCs. Compared with control group, the IL - 6 in POLY group was increased significantly （ P 〈 0.05 ）, and was more in 0.025 g/L group than that in 0.0025 g/L group （ P 〈0.01 ）. The iL- 10 was decreased significantly（P 〈0.01 ） ,and was less in 0.25 g/L group than that in 0. 025 g/L group （P 〈 0.05 ）. However, the IL- 12 was not affected. Compared with the control, the IL -6 in LPS group was increased significantly（P 〈0.01 ） ,while the IL - 10 and IL - 12 were not affected. Conclusion： The TLR3 and TLR4 mRNA are over - expressed in BM - MSCs of healthy donors. The secretion of IL - 10 and IL - 6 in BM - MSCs could be affected by TLR3 activation, while that of IL - 6 could be affected by TLR4 activation. The secretion of IL - 12 in BM - MSCs is not affected by TLR3 or TLR 4 activation.
