| 摘 要： | 目的:检测受体O型蛋白酪氨酸磷酸酶(protein tyrosine phosphatase receptor-type O,PTPRO)在人类肝细胞肝癌(hepatocellular carcinoma,HCC)中的表达水平,研究该基因的表达是否具有性别差异,并是否与HCC肝切除术后肿瘤复发相关,是否可以作为判断预后的指标?方法:①利用实时荧光定量聚合酶链反应和免疫组织化学的方法,分别检测59例男性及22例女性HCC患者肿瘤组织及癌旁组织中PTPRO及雌激素受体(estrogen receptors,ERs)的表达水平;②通过t检验分析PTPRO?ERs在肿瘤?瘤旁及不同性别间的表达差异,并通过相关性分析及F检验了解PTPRO表达与ERs是否具有相关性;③通过Kaplan-Meier曲线研究不同PTPRO表达水平与HCC患者术后复发率之间的关系?结果:①PTPRO?ER?琢在HCC组织中的mRNA及蛋白水平明显低于癌旁肝硬化组织(P < 0.000 1);②HCC癌旁组织中PTPRO?ER?琢的表达水平,男性显著低于女性(P < 0.05);③Kaplan-Meier曲线及Log-rank检验表明HCC癌旁组织中PTPRO水平与HCC复发水平呈负相关(P < 0.000 1)?结论:PTPRO是一种具有性别差异性的抑癌基因,可作为判断HCC肝切除术后预后的指标?
