Smac基因诱导前列腺癌细胞凋亡的实验研究

摘    要:目的:观察Smac基因对前列腺癌细胞凋亡的影响。方法:前列腺癌细胞株(PC-3)在含小牛血清的DMEM-F12培养基中培养、传代。通过脂质体介导将Smac基因导入前列腺癌细胞株,通过RT-PCR法检测SmacmRNA的表达,同时用SABC免疫组化法分析Smac的表达。采用细胞计数法检测细胞生长抑制率,流式细胞仪观察细胞凋亡情况。结果:转染Smac基因后,RT-PCR可检测出Smac的特异条带,SABC结果转染组Smae蛋白表达显著性增强,前列腺癌细胞的生长明显受抑制(P〈0.05)。细胞周期分析可见凋亡峰,凋亡率35.2%。结论:转入Smac基因可显著促进前列腺癌细胞细胞的凋亡。

Abstract:Objective:Present study aimed to observe the effects of Smac gene on killing prostate carcinoma PC- 3 cells. Method.. Prostate carcinoma cells strain, PC-3, was cultured and passaged in DMEM-F12 medium contai- ning bovine serum. Smac gene was transfected into PC-3 cells by liposome. The expression of Smac mRNA was detected using Reverse Transcription Polymerase Chain Reaction(RT-PCR). The expression of protein of Smac was detected by SABC immunohistochemistry. The growth rate of PC-3 cells was studied by constructing the growth curve and calculated as the formula: inhibitory rate= 1-cell number in experiment group/cell number in control group× 100%. Cellular apoptosis was determined by flow cytometry. Result: The RT-PCR result showed the specific band. The difference in SABC-positive cell number between transfected-group and control group was statistically significant ( P 〈0.05). The growth of PC-3 cells was significantly inhibited in transfected-group com- pared with control group ( P 〈0.05). Apoptosis cells increased significantly. The apoptosis rate was 35.2%. Conclusion: Smac gene could inhibit the growth of PC-3 cells effectively.
