反义IGF-1RNA基因治疗脑胶质瘤的实验研究

摘    要:目的 构建反义胰岛素样生长因子-1(IGF-1)表达质粒(pcEP4-IGF-1A)治疗大鼠C6脑胶质瘤。方法 RT-PCR扩增300bpIGF-1 cDNA,测序,构建反义IGF-1表达质粒,体外转染C6细胞,观察其体外增殖速率及对C6胶质瘤的治疗作用。结果 反义IGF-1序列正确,反义IGF-1A转染的C6细胞生长明显受抑,体内成瘤性消失,荷瘤鼠肿瘤全部消失,6个月观察期内肿瘤无复发。结论 反义IGF-1表达质粒能抑制C6细胞体外生长,体内杀伤肿瘤效果明显。

Abstract:Objective To construct expressing plasmid antisense IGF-1 for gene therapy of rat glioma. Methods IGF-1 cDNA 300 bp were amplified using RT-PCR, sequence. Transfect C6 cells with antisense IGF-1 and the cells proliferative rate and the effect on treatment of C6 glioma were ob- served in vivo. Results The sequence was correct. The growth of C6 cells transfected was inhibited significantly and tumorigenicity was lost. After treatment, complete disappearance of tumor occured in all tumor-bearing rats, and no recurrence during six months. Conclusion Antisense IGF-1 expression plasmid inhibits the growth of C6 cells in vitro, and the treatment is effective in vivo.
