摘    要:本实验观察了单克隆抗体55及79对人普通型急性淋巴母细胞性白血病抗原(CALLA)的特异性调变作用,以及抗体浓度、孵育温度及时间对调变作用的影响。此外,发现不仅非T、非B淋巴母细胞Nalm-1可发生CALLA调变,T细胞HPB-ALL,B细胞Daudi及正常人外周血多形核细胞表面CALLA也可发生调变。调变后的Nalm-1细胞对抗CALLA抗体依赖补体的细胞毒作用有明显抵抗力。本文对抗原调变的生物学意义也进行了讨论。


ANTIGENIC MODULATION INDUCED BY MONOCLONAL ANTIBODIES IN VITRO
Abstract:Two monoclonal antibodies 55,79 against human acute lymphoblastic leukemia antigen (CALLA) were used to induce antigenic modulation in this study, A specific loss of GALLA was induced by incubation of Nalm-1 cells with monoclonal antibodies 55/79. It Was found that the CALLA modulation depended on temperature, incubation time as Well as the anybody's concentrations employed for antigenic modulation. In addition, the CALLA modulation observed was not only on Nalm-1 cell but, to certain extent, also on Daudi, HPB-ALL and polymorphs. The loss of OALLA on Nalm-1 cells inhibited the complement-dependent cytotoxicity of McAb 55/79. The results suggested that the CALLA modulation might play a role in certain pathological conditions.
