术前区域动脉灌注5-Fu联合奥沙利铂对于结肠癌细胞增殖和凋亡的影响及其机制研究

摘    要:目的:研究术前区域动脉灌注5-Fu和奥沙利铂对于结肠癌细胞增殖和凋亡的影响。方法:选择在本院确诊为结肠癌的60例患者,随机分为5-Fu联合奥沙利铂组(A组)、5-Fu组(B组)、奥沙利铂组(C组)、对照组(D组),每组15例。分别于术前区域动脉灌注相应的药物。检测各组肿瘤组织中Ki标记指数(Ki67LI)、凋亡细胞数目、凋亡指数(AL)、凋亡基因含量。结果:A组肿瘤组织中Ki标记指数明显低于B组、C组以及D组;A组肿瘤组织中的凋亡细胞数目和凋亡指数明显低于B组、C组以及D组;A组肿瘤组织中凋亡基因Caspase-3、Caspase-9、Fas、FasL的蛋白质表达量均高于B组、C组以及D组。以上差异均具有统计学意义。结论:术前区域动脉灌注5-Fu和奥沙利铂能够显著性抑制结肠癌肿瘤细胞的增殖,促进肿瘤细胞的凋亡,这种作用的可能机制是上调Caspase-3、Caspase-9、Fas、FasL的蛋白质表达量。

Effects of preoperative regional arterial infusion of 5-Fu combined with oxaliplatin on colon cancer cell proliferation and apoptosis and its mechanism
Abstract:[]Objective To study the effect of preoperative regional arterial infusion of 5-Fu and oxaliplatin on colon can-cer cell proliferation and apoptosis. Methods 60 cases of colon cancer patients who diagnosed in our hospital were randomly divided into 5-Fu combined with oxaliplatin group(A), 5-Fu group(group B), oxaliplatin group(group C)the control group(group D), 15 cases each. Respectively infused corresponding drug in preoperative regional artery. The tumor tissue Ki labelingindex(Ki67LI), number of apoptotic cells and apoptotic index(AL), and apoptosis gene content were detected. Results The Ki67LI, number of apoptotic cells and AL in group A was lower than that in others groups;the apoptotic gene Caspase-3, Cas-pase-9, Fas and FasL protein expression in group A were higher than those in other groups. Conclusion Preoperative arterial infusion of 5-Fu and oxaliplatin can significantly inhibit the proliferation of colon cancer cells and promote apoptosis of tumor cells, the mechanism of this effect is upregulating Caspase-3, Caspase-9, Fas and FasLprotein expression.
