黄芪甲苷对人膝骨关节炎退变软骨细胞MMP-1和MMP-3 mRNA表达的影响

摘    要:目的:观察黄芪甲苷对原代培养骨关节炎患者的膝关节软骨细胞的金属蛋白酶-1和金属蛋白酶-3mRNA表达影响,初步探讨黄芪甲苷防治膝骨关节软骨退变的作用机制。方法:原代培养骨关节炎患者的膝关节软骨细胞,用第三代细胞进行试验。将5个不同浓度的黄芪甲苷,分别作用于关节软骨细胞4、8、24、48、72h后,采用Real Time-PCR法检测细胞中金属蛋白酶-1(MMP-1)和金属蛋白酶-3(MMP-3)的mRNA表达。结果:黄芪甲苷浓度为25~100mmol/L,作用于退变关节软骨细胞24~72h时,软骨细胞MMP-1 mRNA的表达量与对照组比较显著降低。黄芪甲苷浓度为50~100mmol/L,作用于退变关节软骨细胞48~72h时,软骨细胞MMP-3 mRNA的表达量显著低于对照组。结论:黄芪甲苷可延缓膝骨关节炎关节软骨细胞的退变,抑制软骨细胞MMP-1和MMP-3mRNA的表达可能是其作用机制之一。

Abstract:Objective:The aim of this study was to investigate the effects of astragaloside IV on the expressions of MMP-1and MMP-3 mRNA in chondrocytes of patients with osteoarthritis.Methods:Chondrocytes from the resected knee joint cartilage of patients with OA were isolated and cultured.The chondrocytes of third generation were incubated with astragaloside IV for 4,8,24,48,72h.The expressions of MMP-1 and MMP-3 mRNA in chondrocytes were measured by real-time polymerase chain reaction(RT-PCR).Results:Astragaloside IV decreased the MMP-1and MMP-3 messenger RNA expression levels in chondrocytes of patients with osteoarthritis.Conclusion:The results suggest that astragaloside IV may protect cartilage in the development of OA.Inhibition of the expressions of MMP-1 and MMP-3 mRNA was one of the mechanism of astragaloside IV protected cartilage.
