射干中多种异黄酮类化合物HPLC检测含量研究方法的建立

摘    要:目的:建立HPLC法测定射干中多种异黄酮类化合物含量的测定方法。方法:采用乙腈-2%磷酸水溶液梯度洗脱,流速:1.0mL/min,进样量:10μL,检测波长:266nm,柱温:25℃,用外标法定量,同时测定中鸢尾苷、野鸢尾苷、鸢尾黄素、野鸢尾黄素、次野鸢尾黄素和白射干素等6种异黄酮类成分的含量。结果:经过HPLC法测定不同产地射干药材的含量以及在根茎与其须根的含量上进行比较,发现彼此之间含量具有很大差异,通过本次实验所得到的各成分含量数值作为参数,能够准确、真实、直观地揭示HPLC法对建立10个不同产地射干药材及须根中多种异黄酮类化合物成分的质量控制的标准性。结论:对不同产地射干中6种异黄酮类成分含量测定结果显示,此法可用于射干药材中6种异黄酮类化合物成分含量的同时测定。

Abstract:Objective:Established by reveed-phase HPLC Blackberrylily Rhizome multiple isoflavone compounds content determination of method.Methods:Using acetonitrile-2% phosphoric acid aqueous gradient elution,Flow rate:1.0mL/min,sampling volume:10μL,detection wavelength:266nm,Column temperature:25℃,with external standard method quantitative and simultaneous determination of iridin,Irisflorentin,tectorigenin,irigenin,irisflorentin and dichotomitin six species of isoflavones class concentrations,Result:After by reveed-phase HPLC different placesof Blackberrylily Rhizome medicine content and its content in rhizome regarding the comparison,find each other content has very big difference,through this fokker-planck equation of each component content numerical as parameters,to accurately,real and intuitive to reveal the HPLC to establish 10 different origin Blackberrylily Rhizome medicine and the variety of isoflavones compounds in regarding quality control standard component.Conclusion:Among the different placesof shoot dry ingredients like in 6 kinds of isoflavones content determination results show that this method can be used to Blackberrylily Rhizome medical 6 kinds of isoflavones compounds component content of simultaneous determination.
