摘 要: | 目的:研究摄入次硫酸氢钠甲醛后小鼠大脑皮层中Caspase-3、NOS-1表达的变化,以探讨次硫酸氢钠甲醛对大脑的影响。方法:健康雄性昆明小鼠,随机分为10d组、30d组,每组再分为对照组和0.4‰、0.8‰、1‰三个剂量实验组。10d后处死10d组小鼠;30d后处死30d组小鼠,取出大脑,采用Zamboni固定液固定,常规冰冻制片,免疫组织化学染色,光镜观察。结果:10d实验组小鼠大脑皮层中Caspase-3、NOS-1阳性表达同对照组比较表达明显增强,不同剂量间未见显著性差异。30d实验组小鼠大脑皮层中Caspase-3阳性表达和对照组相比无显著性变化,不同剂量间未见明显差异;30d实验组小鼠大脑皮层中NOS-1阳性表达同对照组相比明显降低,不同剂量间未见显著性差异。结论:次硫酸氢钠甲醛喂食后,可引起小鼠大脑皮层中Caspase-3、NOS-1表达增高,提示次硫酸氢钠甲醛促使大脑皮层中神经元凋亡增加,长期摄入可使神经元濒临死亡。

Effects of formaldehyde sulphoxylate on the expression of Caspase-3 and NOS-1 in mouse brain |

Abstract: | Objective:To study the changes of the expression of Caspase-3 and NOS-1 in mouse brain and discuss the effect of formaldehyde sulphoxylate on mouse brain. Methods: Take healthy male mice of Kunming genus, divide them into two groups at random: 10d group and 30d group. Each group is separated into control group, 0.4‰,0.8‰ and 1‰ tests group. On the 10th day, quickly take out mice brains in 10d group; On the 30th day, quickly take out mice brains in 30d group; fixed in the Zamboni fixation solution, conventionally produce frozen section and stain section by immunohistochemical technology. Results: The expression of Caspase-3 and NOS-1 in 10d test group is increased, there is no obvious change in different dose groups. The expression of Caspase-3 in 30d test group is like as control group, there is no obvious change in different dose groups; The expression of NOS-1 in 30d test group is induced, there is no obvious change in different dose groups. Conclusion: After mice was fed Formaldehyde sulphoxylate solution, the expression of Caspase-3 and NOS-1 visibly increased, then induced. Which suggested that formaldehyde sulphoxylate can promote neuronal apoptosis. |
