蝮蛇蛇毒PCA对实验性心肌梗死大鼠血凝状态的影响

摘    要:目的:探讨蝮蛇蛇毒蛋白C激活剂(PCA)对心肌梗死发生后血液凝固性的影响及其机制。方法:SD大鼠24只,随机分为对照组、心肌梗死模型组和PCA实验组。动物麻醉后,从心尖部向左心室注射月桂酸钠(1.0mg/kg)复制心肌梗死(AMI)模型。实验组大鼠于造模前由舌下静脉注射PCA(1.0mg/kg)预处理,由Medlab生物信号采集系统监测心电变化。于造模后30min从颈总动脉取血样2ml,以TEG5000型血栓弹力仪测量各样本血液的凝血时间、血凝块形成时间、Alpha角度、凝血最大幅度和凝血指数等参数。结果:模型组大鼠血样的R值较对照组明显缩短(P<0.01),CI值>+3.0,而Alpha角度变化不大;LY30%值明显增大,但小于7.5%。PCA实验组大鼠血样的R值较模型组明显延长,MA值减小(P<0.05),LY30%、A值、CI值明显减少(P<0.01)。结论:心肌梗死大鼠血液表现显著高凝状态,蛇毒PCA组分的预处理可以减轻这一过程。

Abstract:Objective:To investigate the effects and mechanisms of protein C activator (PCA) from the venom of agkistrodon halys on blood coagulation after the myocardial infarction. Methods:Twenty-four SD rats were randomly divided into control,myocardial infarction(AMI) or protein C activator (PCA) group. After anesthesia,the rats were given injection of sodium laurate (1.0 mg / kg) by the apex into left ventricle for replicating the model of acute myocardial infarction(AMI). PCA group were administered PCA(1.0 mg/kg) by sublingual intravenous injection before model replication,with ECG monitoring by Medlab bio-signal acquisition system. Thirty minutes after the animal models were prepared,blood samples of 2 ml were taken the from carotid arteries for measuring the blood with regard to time of coagulation reaction and formation,rate of blood coagulation speed(alpha),coagulation strength and index by thromboelastography TEG5000(Haemoscope,US). Results: The R value in AMI group reduced significantly as compared with the controls (P<0.01)and CI values were greater than +3.0,but the Alpha angle changed little with the MA value. LY30% was significantly increased,but was less than 7.5%. PCA group had much longer R value of blood than the model group,with reduced MA value(P<0.05),LY30%,A value and CI (P<0.01).Conclusion: Rats with AMI are characterized by hypercoagulability,but the findings suggest that venom PCA could reduce the incidence of the coagulation.
