HBx蛋白表达和ROS水平与肝细胞癌的关系及其意义

摘要] 目的 探讨乙型肝炎病毒X （HBx）蛋白表达和活性氧（ROS）水平与肝细胞癌的关系及其意义。方法 选取肝细胞性肝癌（肝癌）标本42例为实验对象，每例标本均选取癌和癌旁组织；同时选取肝外伤或肝血管瘤正常肝组织标本20例为正常对照组。采用SABC法检测组织中的HBx蛋白，用流式细胞仪检测ROS水平。结果 HBx蛋白在正常肝组织中阴性；在癌及癌旁组织中的阳性例数分别为31例（73.8%）和35例（83.3%），差异无显著性（P＞0.05）；肝癌组织细胞内ROS荧光强度明显高于癌旁组织和正常组织(P<0.05)，癌旁组织细胞内ROS荧光强度明显高于正常组织（P<0.05）；癌组织和癌旁组织中HBx蛋白（+）者的ROS水平均明显高于HBx蛋白（-）者（P＜0.05）。结论 HBx蛋白的表达与ROS水平明显相关，HBx可能通过ROS的积累导致肝癌的发生。

The relationship and the evaluation among the expression of HBx protein,the level of ROS and hepatocellular carcinoma

Abstract]Objective To explore the relationship and the evaluation among the expression of HBx protein ,the level of ROS and hepatocellular carcinoma(HCC).Methods 42 HCCs were selected as study groups,and the tumor tissues and the adjacent-tumor tissues were all studied.20 cases who were liver surgical trauma patients or hemangiomas of liver were selected as normal control group. The expression of the HBx protein was detected by SABC immunohistochemistry and the levels of ROS were measured by flow cytometry.Results The expression of the HBx protein was negative in normal liver tissues while there were 31 cases（73.8%）in tumor tissues and 35 cases（83.3%）in adjacent-tumor tissues positive without significantly difference(P＞0.05). The levels of ROS in HCC tissues were significantly higher than that in the adjacent-tumor tissues（P＜0.05）,and the similarly result existed between the adjacent-tumor tissues and the normal liver tissues（P＜0.05）.The levels of ROS in tumor tissues whose HBx protein expression was positive was higher than that whose HBx protein expression was negative(P＜0.05),and the similarly result existed in the adjacent-tumor tissues(P＜0.05).Conclusion There was a relationship between the expression of the HBx protein and the level of ROS,and HBx might cause HCC via accumulating the ROS.
