小鼠白血病Cyclin G和其它相关蛋白表达及凋亡研究

摘    要:目的:探讨在白血病细胞中细胞周期素zCyclin)G,Cyclin D,P16,P21,P53等基因的蛋白表达水平和白血病细胞周期变化。方法:建立白血病模型(L615);采用免疫组织化学法检测小鼠的Cyclin G、P53、P21、Cyclin D、P16和C-fos、bcl-2的表达;流式细胞术检测小鼠白血病细胞的凋亡率和细胞周期。结果;阳性组和复发组的Cyclin G、Cyclin D、C-fos表达明显高于正常组和化疗组(P<0.05),P16则相反(P<0.01);P21在四组中的表达差异无显著性意义(P>0.05);阳性组和复发组P53的表达为阴性,而化疗组与正常组表达差异无显著性意义(P>0.05);bcl-2四组表达差异无显著性意义(P>0.05);化疗组和阳性组的细胞凋亡率均较正常组增高(P<0.05),且化疗组的凋亡率比阳性组的明显增高(P<0.01)。结论:白血病的发生、发展可能与Cyclin G、Cyclin D、C-fos基因的高表达,P16低表达有关。

Abstract:Objective:To study expression of Cyclin G?P53?P21?Cyclin D?P16 and C-fos ?bcl-2 in leukemia mice. Method:By established mice models with leukemia (L615); the protein expression of Cyclin G?P53?P21?Cyclin D?P16 and C-fos ?bcl-2 were measured with immunohistochemical method;the rate of apototic cells and cell cycle were detected by flow cytometry.Result:Expression of Cyclin G?Cyclin D?C-fos in control group and relapse group was higher than that in normal group and chemotherapy group (P< 0.05) , expression of P16 was reverse(P< 0.01); expression of P21 showed no difference(P> 0.05), expression of P53 in control group and relapse group was negative , in normal group and chemotherapy group showed no difference(P< 0.05) , expression of bcl-2 showed no difference(P> 0.05) ; the rate of apoptotic leukemia cell in chemotherapy group and control group was higher than that in normal group(P< 0.05) , and chemotherapy group was obviously higher than normal group(P< 0.01).Conclusion:Leukemia is related to higher expression of Cyclin G?Cyclin D?C-fos gene and lower expression of P16, the rate of apoptosis of leukemia cell can be induced by chemotherapy.
