血管内皮生长因子及其受体在食管鳞癌组织中的表达及意义

摘    要:目的 :检测了食管鳞癌组织中血管内皮生长因子 ( VEGF)及其受体 ( Flk- 1 )的表达、分布。探讨二者在食管鳞癌浸润转移中的作用及临床意义。方法 :采用链霉菌抗生素蛋白 -过氧化物酶 ( S- P)免疫组织化学法检测 1 6例正常食管粘膜 ,45例食管鳞癌石蜡切片中 VEGF及 Flk- 1的表达及分布情况。结果 :45例食管鳞癌组织 ,VEGF阳性表达主要集中于癌细胞胞浆中 ,Flk- 1阳性表达主要在癌组织的血管内皮细胞及癌细胞中。正常食管粘膜仅 1例 VEGF弱阳性表达。VEGF及 Flk- 1阳性表达随食管鳞癌的临床分期升高 ,二者均有升高 ,且二者与强度等级显著相关 ( P<0 .0 5 )。有淋巴结转移的癌组织中 VEGF及 Flk- 1的表达明显高于无淋巴结转移的癌组织 ( P<0 .0 5 )。结论 :VEGF及 Flk- 1的表达与食管癌浸润、转移关系密切。VEGF及 Flk- 1可作为临床评价疗效及预后的一项重要参考指标

Abstract:Objective:To at observe the expression of VEGF and Flk-1 in esophageal carcinoma tissues and to analyze its effects and significance in tumor invasion and metastasis. Methods: VEGF and Flk-1 expressions were detected from 16 cases of normal esophageal mucosas and 45 cases of esophageal carcinoma samples with streptavidin-biotin-HRP(s-p) immunohisto-chemical method. Results: VEGF is mainly expressed in cytoplasm of cancer cells, while Flk-1 expressed in both cancer cells and vascular endothelial cells. One case of normal esophageal mucosas shows feeble expression of VEGF.The more advanced the clinical stage of squamous carcinoma,the higher rate of VEGP and Flk-1 expression (P<0.05). Besides, the expression of VEGF and Flk-1 in esophageal carcinoma tissues with lymph nodes metastasis is higher than that of esophageal carcinoma tissues without lymph nodes metastasis (P<0.05). Conclusion: The positive expressions of VEGF and Flk-1 have a significant relationship with invasion and metastasis in esophageal carcinoma. The level of VEGF and Flk-1 expression may provide a valuable index for prognosis and efficacy of clinical treatment in esophageal carcinoma.
