转染 p53基因对肺腺癌细胞株裸鼠 移植瘤生长的影响

摘    要:目的:探讨转染野生型 p53( wt-p53)和突变型 p53( mt-p53)基因对人肺腺癌细胞株 GLC-82裸鼠移植瘤生长的影响。方法:采用脂质体介导法,分别将 wt-p53和 mt-p53基因导入人肺腺癌细胞株 GLC-82,在裸鼠体内、体外实验中检测转导细胞的生长状况和裸鼠致瘤性。结果:转染 mt-p53 基因的细胞株 G418筛选的细胞集落数、 3H-TDR掺入实验、软琼脂平皿细胞集落数,以及裸鼠瘤组织重量和体积均高于对照组( P<0.01),而转染 wt p53基因的细胞株均显著低于对照组( P< 0.01),表明导入 wt p53基因的细胞株瘤细胞生长速度明显低于对照组细胞株和导入 mt p53基因的细胞株,即导入 mt p53基因的细胞株瘤细胞生长速度最快,而导入 wt p53基因的细胞株瘤细胞生长速度最慢。结论: wt p53基因能有效抑制人肺腺癌细胞生长; mt p53基因则可以明显地促进瘤细胞生长。

Abstract:Objective:This study was designed to explore th e significance and the role of wild-type p53(wt-p53)gene and mutant p53gene(mt-p53)transfer on human glandular lung can cer cell growth in nude mice.Methods :wt-p53gene and mt-p53gene were tran sfected and lipofectin-mediated in to the human glandular lung cancer ce ll line GLC-82.And the growth of gene-transfected cell lines were observed in vitro and in vivo.Results:The colony number in the colong-forming experiment and the volume and weight in nude mice were greater in the mf-p53tranfecting cells group than in the c ontrol group.The tumor resulting fr om the cells transfected with the wt-p53gene grew more slowly and was smaller than that from control GLC-82cells.In contra st,the tumor from the cells transfec ted with the mt-p53gene grew faster than that pro duced by cells transfeted with the wt -p53gene and that produced by contro l GLC-82cells.Conclusion:The wild-type p53gene could inhibit the glandular lung cancer cell growt h in nude mice and the mutant p53gene could enhance the glandular lung cancer cell growth in nude mice.
