臭牡丹对神经病理性痛机械痛敏和脊髓背角谷氨酸的作用

摘    要:目的:探讨臭牡丹对神经病理性痛大鼠的机械痛敏和脊髓背角谷氨酸含量的作用。方法:选择神经病理性疼痛坐骨神经分支选择损伤(SNI) 大鼠模型,实验随机分为4组:对照组(腹腔注射生理盐水)、假手术组(分离坐骨神经但不剪断+腹腔注射生理盐水)、模型组(坐骨神经剪断+腹腔注射生理盐水)、实验组(坐骨神经剪断+腹腔注射臭牡丹30 g·kg-1)。运用机械缩足反射阈值(MWT)检测机械痛敏,高效液相法测定大鼠脊髓背角中谷氨酸的含量。结果:与模型组比较,给予臭牡丹组后第7、10、14、21天的机械缩足反射阈值(MWT)明显升高(P<0.05,P〈0.001),在第7天及21天观察到臭牡丹可显著降低大鼠脊髓背角谷氨酸的含量(P<0.001)。结论:臭牡丹可减轻神经病理性痛大鼠的机械痛敏,其机制可能与抑制脊髓背角谷氨酸的表达上调有关。

Abstract:Objective: To explore the effect of clerodendron bungei steud on the mechanical allodynia and glutamate contents in spinal cord in rats with neuropathic pain. Methods: The model of spared nerve injury induced neuropathic pain( SNI) was selected in this experiment. Rats were randomly divided into 4 groups: control group,sham operation group,model group( SNI) and experimental group( clerodendron bungei steud treatment group). Mechanical withdrawal threshold( MWT) was measured as the sign of Mechanical allodynia. The lumbar segment of the spinal cord was removed for determination of glutamate contents by high efficiency liquid chromatography( HPLC). Results: Compared with control group,the mechanical withdrawal threshold( MWT) of clerodendrum group was significantly higher( P 0. 05,P 0. 001) on 7th,10 th,14th,and 21 st days. Clerodendrum observed on the 7th day and 21-day could significantly reduce the content of glutamic acid in Rat Spinal dorsal horn( P 0. 001). Conclusion: Clerodendron bungei steud can relieve the mechanical allodynia in neuropathic pain; its mechanism may be relevant to inhibit the expression of glutamate in spinal dorsal horn.
