摘    要:目的观察丹参多酚酸盐对大鼠局灶性脑缺血再灌注损伤后脑组织内脑源性神经营养因子(BDNF)及胶质源性神经营养因子(GDNF)表达的影响,探讨其神经保护作用的机制。方法线栓法制作大鼠大脑中动脉闭塞再灌注损伤模型,SD大鼠随机分为正常对照组、缺血再灌注组、丹参多酚酸盐低剂量治疗组(10mg/kg)和丹参多酚酸盐高剂量治疗组(30mg/kg),观察各组大鼠的行为学改变,测定脑梗死体积,ELISA法检测脑组织中BDNF和GDNF的含量。结果与缺血再灌组比较,丹参多酚酸盐可以显著降低大鼠神经损伤行为学评分,减少脑梗死体积,增加脑组织内BDNF和GDNF的含量,且呈剂量依赖性。结论丹参多酚酸盐对脑缺血再灌注损伤有明显的保护作用,其机制与促进脑组织合成BDNF及GDNF有关。

The effect of salvianolate on the expresson of BDNF and GDNF in focal cerebral ischemia-reperfusion rats

Abstract:Objective To explore the neuroprotective effect of salvianolate on the expression of BDNF and GDNF in focal cerebral ischemia-reperfusion rats. Methods Longa method was used to establish the MCAO model. SD rats were randomly divided into four groups: sham control group, ischemia-reperfusion group, low dose salvianolate group (10mg/kg) and high dose salvianolate group (30mg/kg). The salvianohe was injected intraperitoneally at designated time point. The cerebral infarction volume was measured by TTC stain and the behavior score was detemained by Longa's method. The expression of BDNF and GDNF in brain tissue was tested by ELISA respectively. Results Compared with ischemia control group, salvianolate could obviously reduce the cerebral infarction volume and the behavior score by upregulating the expression of BDNF and GDNF in brain tissue which indicating the neuroprotective effect of salvianolate in ischemia damage. Fur- thermore, the salvianolate neuroprotective effect was much higher at the high dose than that at the low dose. Conclusions Salvianolate is neuroprotective in ischemia damage by upregulating the expressions of BDNF and GDNF in brain tissue in a dose-dependent manner.
